    Abstract: In Brazil, prostate cancer is a common disease among men. The radionuclide therapy with PSMA analogs, labeled with beta and alpha emitters, has brought new perspectives for patients with multi metastatic resistant prostate cancer. The commercialization of radiopharmaceutical becomes a challenge to transport and ensure the quality of the product, especially in terms of radiochemical stability of therapeutic radiopharmaceuticals. The effect of radiolysis on therapeutic doses of radiopharmaceuticals can be avoided or mitigated by diluting the final dose and reducing storage and transportation temperature. This study evaluated the effect of radiolysis on the different radioactive concentration in the industrial batches of 177Lu-PSMA-617 and in the fractionated doses, considering long time transportation. The radiopharmaceutical 177Lu-PSMA-617 was produced in batches reaching 37 GBq, and stability studies were performed in controlled conditions. The results showed that a combination of factors, including reaction buffer, the radioactive concentration of final product, and freezing storage contributed to the stability of the radiopharmaceutical for 48 hours, enabling transport of 177Lu-PSMA-617 to distant regions of the country.

    Abstract: The World Health Organization (WHO) estimates that the number of new cancer cases worldwide will reach 15 million by 2020. The disease is already the second leading cause of death worldwide, being behind only cardiovascular disease. It is unquestionable that it is a public health problem, especially among developing countries. Prostate cancer is the most common among men, approximately 28.6%. The choice of type of treatment for prostate cancer should consider several factors such as: tumor size and extent, apparent aggressiveness (pathological characteristics), age, health. Among the methods applied, brachytherapy has been used in the initial and intermediate stages of the disease. Brachytherapy is a safe and effective treatment for localized prostate cancer. Brachytherapy is a form of radiotherapy in which radioactive seeds are placed in contact with or within the organ being treated. This technique allows a large dose of radiation to be released only on the target tumor that protects healthy surrounding tissues. Sources may have different shapes and sizes, but the one used for prostate cancer is usually 4.5 mm in length and 0.8 mm in diameter. About 80 to 120 seeds can be used per patient. Iodine-125 is the radioisotope most used in brachytherapy of the prostate, it emits 35,49keV X-rays in 100% of the decays, with average energy of 29 keV. The treatment of prostate cancer with permanent implantation of iodine-125 seeds has grown dramatically in the world in recent years. Most patients can return to normal life within three days with little or no pain.

    Abstract: Three-dimensional (3D) cell culture is increasingly being used in assays to assess the safety and efficacy of new drug candidates. Tumor cell spheroids can mimic with high precision the biological complexity of cellular interactions with their tumor microenvironment. Currently, several techniques can be used to construct 3D spheroids. Among them, magnetic levitation is one of the most used in biomedical research. This technique consists in the magnetization of cells through the adsorption of magnetic nanoparticles of iron oxide (Fe3O4) that are produced by the reaction of Fe2+ and Fe3+ ions in alkaline medium. In this work, nanoparticles of paramagnetic iron oxide (PIONS) were synthesized by coprecipitation through electron beam irradiation at 15 and 30 kGy doses. After functionalization with polar amino acids, nanoparticle suspensions were characterized by physical-chemical assays that showed the successful attachment of the carboxylate groups to the iron, explaining the ability of the particles to adsorb the membranes. Cytotoxicity assay showed that the nanoparticles synthesized by microwave (MW) and electron beam had no toxicity. Others biological assays have also shown efficient adsorption of the particles by human prostate tumor cells, allowing the in vitro application of a biomimetic 3D biological model with potential utilization regarding the development and evaluation of antitumor drugs and radiopharmaceuticals for the treatment of prostate cancer.

    Abstract: Gaining prominence in clinical practice, the 68Ga, positron emitter radionuclide easily obtained by 68Ge/68Ga generator elution, has shown potential and excellent quality on radiolabeling of peptide for use in positron emission tomography (PET), in particular urea-based inhibitor peptides, directed to the prostate-specific membrane receptor (PSMA). Previous studies with the PSMA linked to the chelator HBED-CC (PSMA-11) radiolabeled with 68Ga showed high contrast PET/CT images to evaluate recurrence and metastasis of prostate cancer, becoming an important imaging agent in the clinical routine. This work intended to evaluate the influence of the quality of the 68Ge/68Ga generator eluate in direct labeling of PSMA-11 with 68Ga, assisting in the development of kit for prompt radiolabeling. It was evaluated the 68GaCl3 eluate from 68Ge/68Ga non-GMP generator (manufacturer A) and 68Ge/68Ga GMP generator (manufacturer B), both commercially available. To evaluate the influence of the 68Ga eluate on radiochemical yield of the preparations, the radiochemical purity was determined by thin layer chromatography and HPLC. The radiolabeling with non-GMP generator eluate was determined with and without preliminary purification of the 68 gallium chloride eluate, employing cationic purification columns. The results showed higher radiochemical yield with the 68GaCl3 eluate from the 68Ge/68Ga GMP generator, obtaining the radiolabeled product more easily and speed to clinical practice, without preliminary purification, as opposed to the use of non-GMP 68Ge/68Ga generator which required preliminary purification of the 68GaCl3 eluate to promote satisfactory radiochemical purity results.

    Abstract: This work evaluates the δ -aminolevulic acid (ALA)-induced Protoporphyrin IX (PpIX) synthesis and the PPIX transference from the tumor cells to the whole blood cells. Human prostate tumor cells (DU145 cells) received ALA at different periods and doses to analyze the PPIX synthesis. Thereafter, blood of healthy animals was added to the cultured tumorigenic cells to monitor the transference of the PPIX synthesized from them to the blood cells. Our results demonstrated that the intracellular production and accumulation of PPIX into prostate human tumor cells depends on the ALA doses and the time of exposition. Besides, PPIX molecules synthesized by tumor cells were transferred to the blood cells, but they were not transferred to the DMEM medium solution, showing that it is not a simple diffusion process of PPIX migration, but probably a complex efflux mechanism that cells have developed to transport it through the blood cells to regulate PPIX concentration inside them, since an excess of PPIX is highly toxic. These findings can be useful to understand the accumulation of PPIX to the blood of tumor-bearing animals and cancer patients, and it can be useful to design treatment and diagnosis protocols.

    Abstract: Introduction: Functionalization of water-soluble chitosan (WSCS) nanocolloids with, gold nanoparticles (AuNPs), and LyslLys3 (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid)-bombesin 1– 14 (DOTA-BBN) peptide affords an innovative pathway to produce prostate tumor cell-specific nanomedicine agents with potential applications in molecular imaging and therapy. Methods: The preparation involves the production and full characterization of water-soluble chitosan (WSCS), via gamma (γ) rays (80 kGy) irradiation, followed by DOTA-BBN conjugation for subsequent use as an effective template toward the synthesis of tumor cell-specific AuNPs-WSCS-DOTA-BBN. Results: The WSCS-DOTA-BBN polymeric nanoparticles (86 ± 2.03 nm) served multiple roles as reducing and stabilizing agents in the overall template synthesis of tumor cell-targeted AuNPs. The AuNPs capped with WSCS and WSCS-DOTA-BBN exhibited average Au-core diameter of 17 ± 8 nm and 20 ± 7 nm with hydrodynamic diameters of 56 ± 1 and 67± 2 nm, respectively. The AuNPs-WSCS-DOTA-BBN showed optimum in vitro stability in biologically relevant solutions. The targeted AuNPs showed selective affinity toward GRP receptors overexpressed in prostate cancer cells (PC-3 and LNCaP). Discussion: The AuNPs-WSCS-DOTA-BBN displayed cytotoxicity effects against PC-3 and LNCaP cancer cells, with concomitant safety toward the HAECs normal cells. The AuNPs-WSCS-DOTA-BBN showed synergistic targeting toward tumor cells with selective cytotoxicity of AuNPs towards PC-3 and LNCaP cells. Our investigations provide compelling evidence that AuNPs functionalized with WSCS-DOTA-BBN is an innovative nanomedicine approach for use in molecular imaging and therapy of GRP receptor-positive tumors. The template synthesis of AuNPs-WSCS-DOTA-BBN serves as an excellent non-radioactive surrogate for the development of the corresponding 198AuNPs theragnostic nanoradiopharmaceutical for use in cancer diagnosis and therapy.

    Abstract: Porphyrin accumulate substantially more in tumors than in normal tissues. The optical nonlinearity of the blood porphyrin was analyzed using Z-scan technique. The results showed a decrease in nonlinear refractive index value for tumor blood.

Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.

A elaboração do projeto do RI do IPEN foi iniciado em novembro de 2013, colocado em operação interna em julho de 2014 e disponibilizado na Internet em junho de 2015. Utiliza o software livre Dspace, desenvolvido pelo Massachusetts Institute of Technology (MIT). Para descrição dos metadados adota o padrão Dublin Core. É compatível com o Protocolo de Arquivos Abertos (OAI) permitindo interoperabilidade com repositórios de âmbito nacional e internacional.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.

Operando, inicialmente como uma base de dados referencial o Repositório foi disponibilizado na atual plataforma, em junho de 2015. No Repositório está disponível o acesso ao conteúdo digital de artigos de periódicos, eventos, nacionais e internacionais, livros, capítulos, dissertações, teses e relatórios técnicos.

O gerenciamento do Repositório está a cargo da Biblioteca do IPEN. Constam neste RI, até o presente momento 20.950 itens que tanto podem ser artigos de periódicos ou de eventos nacionais e internacionais, dissertações e teses, livros, capítulo de livros e relatórios técnicos. Para participar do RI-IPEN é necessário que pelo menos um dos autores tenha vínculo acadêmico ou funcional com o Instituto. Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens.
