Abstract:
Atherosclerosis is a chronic inflammatory disease and the main cause of human death worldwide. Studies have proposed that macrophages play a significant role in the development, progression, and destabilization of atherosclerotic plaques[1]. Thus, reduction of macrophages from plaque represents a new strategy for the treatment of atherosclerosis. Photodynamic and Sonodynamic therapies (PDT and SDT, respectively) are emerging as new atherosclerosis treatments [1, 2]. The subsequent generation of free radicals by activated photo and sonosensitizers can lead to apoptotic cell death. The use of gold nanoparticles (AuNPs) as the vehicle for a sensitizer delivery improves reactive oxygen species formation [3]. Previous works demonstrated that aminolevulinic acid (5-ALA) mediated PDT could reduce macrophage content and inhibit plaque progression, indicating a promising approach to treat inflamed atherosclerotic plaque[4]. In this study ALA:AuNps functionalized with polyethylene glycol (PEG) were synthesized mixing 5-ALA with tetrachloroauric(III) acid in milliQ water solution followed by photo reduction with 300 Watts Xenon lamp. The synthesized nanoparticles were characterized by UV/Vis optical absorption, and electron microscopy. Singlet oxygen generation efficiency was measured by trapping with 1,3-diphenylisobenzofuran for irradiations with 590 nm LED (~100 mW) and pulse ultrasound irradiation (1 and 3W/cm2 with 1.0 and 2.0 MHz ). The potential use of ALA:AuNps as a sensitizer for photo and sonodynamic therapies were investigated on THP-1 macrophage. Cytotoxicity test were also described. The findings suggested that ALA:AuNps under LED illumination and ultrasound had photo and sonodynamic effect on THP-1 macrophages via generation of intracellular singlet oxygen, indicating that ALA:AuNps could be used as a novel photo and sonosensitizer for atherosclerosis.

Abstract:
Photodynamic and sonodynamic therapies (PDT and SDT, respectively) are emerging as new atherosclerosis
treatments. The subsequent generation of free radicals by activated photo and sonosensitizers can lead to
apoptotic cell death. The use of gold nanoparticles (AuNPs) as the vehicle for a sensitizer delivery improves
reactive oxygen species formation and sensitizer performance. In this study gold nanoparticles functionalized
with polyethylene glycol (PEG) were synthesized mixing δ-aminolevulinic acid (ALA) with tetrachloroauric (III)
acid in milliQ water solution followed by photo reduction with 300W xenon lamp. The synthesized ALA:AuNPs
were characterized by UV/vis optical absorption, zeta potential and electron microscopy. The mean particle size
of spherical ALA:AuNP was ~ 18 nm, with a polydispersity index of 0.437. Singlet oxygen generation efficiency
was measured using the trap molecule 1,3-diphenylisobenzofuran. ALA:AuNPs and DPBF were irradiated with
590 nm LED, or pulse ultrasound irradiation (1 W/cm2 with 1.0 MHz), and consumption of the DPBF was
monitored over time by the absorption and emission spectra. The results showed that he gold nanoparticles
generate singlet oxygen during light and ultrasound irradiations. THP-1 cells differentiated into macrophages
cytotoxicity test were described and was found the half maximal inhibitory concentration (IC50) values ~ 36 nM
for ALA:AuNPs. Increase in the fluorescence intensity of PpIX extracted from macrophages incubated with
ALA:AuNPs indicating stable encapsulation of ALA into gold nanoparticles and further conversion to PpIX. The
potential use of ALA:AuNps as a sensitizer for photo and sonodynamic therapies were investigated. ALA:AuNPs
mediated SDT was more effective than PDT. SDT with ALA:AuNPs induced the reduction of macrophage viability
in ~ 87,5% in only 2 min. The mechanism underlying SDT-induced apoptosis involves the generation of singlet
oxygen. The results indicate that ALA:AuNPs can be used as a novel photo and sonosensitizer for atherosclerosis.

Abstract:
The functionalization of graphene oxide (GO) with polyethylene glycol (PEG) has been widely used in drug delivery systems. This nanocomposite exhibits excellent stability in the presence of high concentrations of salts and proteins and shows to be less toxic than its raw form in vitro and in vivo. However, it must be sterilized before use in the medical field and the gamma irradiation shows a promising option for this purpose. Sterilization by ionizing energy through gamma rays, generated by Cobalt-60 self-disintegration, consists in exposing the materials to short electromagnetic waves. The irradiation process provides substantial advantages when compared to thermal and chemical processes such as more precise control of the process, production of products with superior qualities, lower energy consumption and less environmental pollution. In this work the effect of gamma radiation on the structure of GO and GO functionalized com Amino-PEG (GO-PEG-NH2) irradiated with different doses (15, 25, 35 and 50 kGy) and rate dose 7.31 kGy.h-1 was evaluated. The analyses were performed by Fourier-transform infrared spectroscopy (FT-IR) and Raman spectroscopy. The results showed that the methods for the synthesis of GO and GO-PEG-NH2 was effective since there was confirmation of the surface oxidation of materials and functionalization with the PEG-NH2 and the sterilization by gamma radiation does not caused any defects on materials.

✔ O RD disponibiliza um quadro estatístico de produtividade, onde é possível visualizar o número dos trabalhos agrupados por tipo de coleção, a medida que estão sendo depositados no RD.

será aberta uma nova página correspondente à aquele autor – trata-se da página do pesquisador.

Constam neste RI, até o presente momento 20.950 itens que tanto podem ser artigos de periódicos ou de eventos nacionais e internacionais, dissertações e teses, livros, capítulo de livros e relatórios técnicos. Para participar do RI-IPEN é necessário que pelo menos um dos autores tenha vínculo acadêmico ou funcional com o Instituto. Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.
A elaboração do projeto do RI do IPEN foi iniciado em novembro de 2013, colocado em operação interna em julho de 2014 e disponibilizado na Internet em junho de 2015. Utiliza o software livre Dspace, desenvolvido pelo Massachusetts Institute of Technology (MIT). Para descrição dos metadados adota o padrão Dublin Core. É compatível com o Protocolo de Arquivos Abertos (OAI) permitindo interoperabilidade com repositórios de âmbito nacional e internacional.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.
Operando, inicialmente como uma base de dados referencial o Repositório foi disponibilizado na atual plataforma, em junho de 2015.
