Brain STAT5 signaling modulates learning and memory formation.

Abstract:
The signal transducer and activator of transcription 5 (STAT5) is a transcription factor recruited by numerous cytokines.
STAT5 is important for several physiological functions, including body and tissue growth, mammary gland development,
immune system and lipid metabolism. However, the role of STAT5 signaling for brain functions is still poorly investigated,
especially regarding cognitive aspects. Therefore, the objective of the present study was to investigate whether brain STAT5
signaling modulates learning and memory formation. For this purpose, brain-specific STAT5 knockout (STAT5 KO) mice
were studied in well-established memory tests. Initially, we confirmed a robust reduction in STAT5a and STAT5b mRNA
levels in different brain structures of STAT5 KO mice. STAT5 KO mice showed no significant alterations in metabolism,
growth, somatotropic axis and spontaneous locomotor activity. In contrast, brain-specific STAT5 ablation impaired learning
and memory formation in the novel object recognition, Barnes maze and contextual fear conditioning tests. To unravel
possible mechanisms that might underlie the memory deficits of STAT5 KO mice, we assessed neurogenesis in the hippocampus,
but no significant differences were observed between groups. On the other hand, reduced insulin-like growth
factor-1 (IGF-1) mRNA expression was found in the hippocampus and hypothalamus of STAT5 KO mice. These findings
collectively indicate that brain STAT5 signaling is required to attain normal learning and memory. Therefore, STAT5 is an
important downstream cellular mechanism shared by several cytokines to regulate cognitive functions.

Abstract:
Growth hormone (GH) exerts important biological
effects primarily related to growth and metabolism.
However, the role of GH signaling in the brain is still
elusive. To better understand GH functions in the brain, we
mapped the distribution of GH-responsive cells and identified
the receptors involved in GH central effects. For this
purpose, mice received an acute intraperitoneal challenge
with specific ligands of the GH receptor (mouse GH),
prolactin receptor (prolactin) or both receptors (human
GH), and their brains were subsequently processed
immunohistochemically to detect the phosphorylated form
of STAT5 (pSTAT5). GH induced pSTAT5 immunoreactivity
in neurons, but not in astroglial cells of numerous
brain regions, including the cerebral cortex, nucleus
accumbens, hippocampus, septum and amygdala. The most
prominent populations of GH-responsive neurons were
located in hypothalamic areas, including several preoptic
divisions, and the supraoptic, paraventricular, suprachiasmatic,
periventricular, arcuate, ventromedial, dorsomedial,
tuberal, posterior and ventral premammillary nuclei.
Interestingly, many brainstem structures also exhibited
GH-responsive cells. Experiments combining immunohistochemistry
for pSTAT5 and in situ hybridization for GH
and prolactin receptors revealed that human GH induced
pSTAT5 in most, but not all, brain regions through both
prolactin and GH receptors. Additionally, males and females exhibited a similar number of GH-responsive cells
in forebrain structures known to be sexually dimorphic. In
summary, we found GH-responsive cells primarily distributed
in brain regions implicated in neurovegetative,
emotional/motivational and cognitive functions. Our findings
deepen the understanding of GH signaling in the brain
and suggest that central GH signaling is likely more ample
and complex than formerly recognized

Abstract:
The human growth hormone receptor antagonist G120R-hGH precludes dimerization of GH and prolactin
receptors and consequently JAK/STAT signaling. Some modifications in this antagonist resulted in a
drug specific for the GH receptor, called Pegvisomant (Somavert®). However, the original G120R-hGH is
usually synthesized in bacterial cytoplasm as inclusion bodies, not being a commercial product. The
present work describes the synthesis and characterization of G120R-hGH secreted into bacterial periplasm
and obtained with a vector based on a constitutive lambda-PL promoter. This antagonist can be
useful for studies aiming at investigating the effects of a simultaneous inhibition of GH and prolactin
signaling, as a potential anti-tumoral or anti-diabetic compound. G120R-hGH, synthesized using the
W3110 E. coli strain, showed a yield of 1.34 ± 0.24 mg/ml/A600 (~0.79 mg G120R-hGH/g of wet weight
cells) after cultivation at 30 C up to 3 A600 units and induction at 37 C, for 6 h, with final 4.3 ± 0.3 A600. A
laboratory scale purification was carried out using three chromatographic steps with a total yield of 32%,
reaching 98% purity. The obtained protein was characterized by SDS-PAGE, Western Blotting, Mass
spectrometry, RP-HPLC, HPSEC and in vitro proliferation bioassay. The proliferation assay, based on Ba/F3-
LLP cells, shows that G120R-hGH (100 ng/ml) significantly inhibited (64%) the proliferative action of hGH
(1 ng/ml). This is the first time that G120R-hGH is synthesized in bacterial periplasmic space and
therefore correctly folded, without the initial methionine. The reasons for a divergent efficacy for
antagonizing hGH versus hPRL is currently unknown and deserves further investigation.

Abstract:
Growth hormone (GH) responsive neurons are extensively distributed in many hypothalamic nuclei that also have leptin receptor (LepR)-expressing cells (1).
However, whether GH affects metabolic functions regulated by leptin remains unknown. In the present study, we initially performed a co-localization study and
confirmed that a large percentage of LepR-expressing neurons are directly responsive to peripherally injected GH in different brain nuclei. Then, we generated
mice lacking GH receptor (GHR) specifically in LepR-expressing cells (LepR GHR KO mice). Although LepR GHR KO mice exhibited a similar body weight,
food intake, energy expenditure, glucose tolerance and leptin sensitivity compared to control mice, we observed a lower adiposity in mutant mice. LepR GHR
KO mice also showed a lower capacity to recover from insulin-induced hypoglycemia and a blunted counterregulatory response evoked by 2-deoxyglucose
(2DG) administration. Co-infusion of 2DG with sympathetic blockers, but not parasympathetic blockers, was able to abolish the differences observed between
groups. Remarkably, while control mice adapted to a 60% food deprivation period by progressively saving energy, LepR GHR KO mice exhibited a blunted
metabolic adaptation to starvation, which led to hypoglycemia and an increased lethality rate, energy expenditure and weight loss, compared to control
animals. In order to identify the specific neuronal populations responsible for the observed responses, we generated mice lacking GHR in steroidogenic
factor-1 (SF1) cells, which comprises the ventromedial nucleus of the hypothalamus (VMH). SF1 GHR KO mice exhibited a similar metabolic phenotype in the
basal condition, compared to littermate controls. On the other hand, SF1 GHR KO mice also showed a lower capacity to recover from insulin-induced
hypoglycemia and a blunted counterregulatory response evoked by 2DG. However, metabolic adaptations to starvation were not affected by SF1-specific GHR
deletion, which suggests that VMH does not mediate these latter changes. In summary, GHR expression in the brain is required to properly regulate glycemia
and energy balance, especially during situations in which GH is highly secreted (e.g., hypoglycemia and food restriction). In addition, our findings revealed a
previously unrecognized role of GH to coordinate, together with leptin, the metabolic adaptations to starvation in order to ensure survival, via the same
neurocircuitry.

Abstract:
The arcuate nucleus (ARH) is an important hypothalamic area for the homeostatic control of feeding
and other metabolic functions. In the ARH, proopiomelanocortin- (POMC) and agouti-related peptide (AgRP)-
expressing neurons play a key role in the central regulation of metabolism. These neurons are influenced by circulating
factors, such as leptin and growth hormone (GH). The objective of the present study was to determine
whether a direct action of GH on ARH neurons regulates the density of POMC and AgRP axonal projections to
major postsynaptic targets. We studied POMC and AgRP axonal projections to the hypothalamic paraventricular
(PVH), lateral (LHA) and dorsomedial (DMH) nuclei in leptin receptor (LepR)-deficient mice (Leprdb/db),
GH-deficient mice (Ghrhrlit/lit) and in mice carrying specific ablations of GH receptor (GHR) either in LepR- or
AgRP-expressing cells. Leprdb/db mice presented reduction in the density of POMC innervation to the PVH compared
to wild-type and Ghrhrlit/lit mice. Additionally, both Leprdb/db and Ghrhrlit/lit mice showed reduced AgRP fiber
density in the PVH, LHA and DMH. LepR GHR knockout mice showed decreased density of POMC innervation in
the PVH and DMH, compared to control mice, whereas a reduction in the density of AgRP innervation was
observed in all areas analyzed. Conversely, AgRP-specific ablation of GHR led to a significant reduction in AgRP
projections to the PVH, LHA and DMH, without affecting POMC innervation. Our findings indicate that GH has
direct trophic effects on the formation of POMC and AgRP axonal projections and provide additional evidence that
GH regulates hypothalamic neurocircuits controlling energy homeostasis.

Abstract:
The quick release bromocriptine mesylate (Cycloset®) was recently approved as a drug to treat type 2 diabetes mellitus, but the exact mechanism of action of this drug remains unknown. Aims: We hypothesized that the anti-diabetic effects of bromocriptine is due to reductions in basal prolactin levels. Furthermore, these effects could be mediated by an interaction with estrogen receptor α (ERα) in the brain. Methodology: Initially, we studied 4 groups of obese and diabetic (ob/ob) mice for 16 days: 1) control; 2) prolactin (micro-osmotic pumps infusing prolactin; 18 μg/day); 3) bromocriptine (one daily ip injection of bromocriptine mesylate; 12 μg/g); and 4) bromocriptine + prolactin. Results: No changes in food intake were observed among the groups; however, bromocriptine groups showed a reduced weight gain along the experiment. Prolactin replacement prevented the improvement in glucose tolerance, insulin sensitivity and serum insulin levels caused by bromocriptine treatment. In another experiment, we found that ERα mRNA showed a very high co-expression with prolactin-responsive cells in hypothalamic nuclei involved in the regulation of glucose homeostasis, such as the arcuate and ventromedial nuclei. Conclusion: Our results suggest that changes in basal prolactin levels are at least partially responsible for the anti-diabetic effects of bromocriptine. These effects are possibly mediated by a crosstalk between prolactin and ERα signaling in the brain. Our findings revealed a novel mechanism that could be target of drugs to prevent and treat diabetes mellitus.

Abstract:
Previous studies have shown that bromocriptine mesylate (Bromo) lowers blood glucose levels in adults with type 2 diabetes mellitus; however, the mechanism of action of the antidiabetic effects of Bromo is unclear. As a dopamine receptor agonist, Bromo can alter brain dopamine activity affecting glucose control, but it also suppresses prolactin (Prl) secretion, and Prl levels modulate glucose homeostasis. Thus, the objective of the current study was to investigate whether Bromo improves insulin sensitivity via inhibition of Prl secretion. Male and female ob/ob animals (a mouse model of obesity and insulin resistance) were treated with Bromo and/or Prl. Bromo-treated ob/ob mice exhibited lower serum Prl concentration, improved glucose and insulin tolerance, and increased insulin sensitivity in the liver and skeletal muscle compared with vehicle-treated mice. Prl replacement in Bromo-treated mice normalized serum Prl concentration without inducing hyperprolactinemia. Importantly, Prl replacement partially reversed the improvements in glucose homeostasis caused by Bromo treatment. The effects of the Prl receptor antagonist G129R-hPrl on glucose homeostasis were also investigated. We found that central G129R-hPrl infusion increased insulin tolerance of male ob/ob mice. In summary, our findings indicate that part of Bromo effects on glucose homeostasis are associated with decrease in serum Prl levels. Because G129R-hPrl treatment also improved the insulin sensitivity of ob/ob mice, pharmacological compounds that inhibit Prl signaling may represent a promising therapeutic approach to control blood glucose levels in individuals with insulin resistance.

✔ Ao efetuar a busca por um autor o RD apresentará uma relação de todos os trabalhos depositados no RD. No lado direito da tela são apresentados os coautores com o número de trabalhos produzidos em conjunto bem como os assuntos abordados e os respectivos anos de publicação agrupados.
✔ O RD disponibiliza um quadro estatístico de produtividade, onde é possível visualizar o número dos trabalhos agrupados por tipo de coleção, a medida que estão sendo depositados no RD.
✔ Na página inicial nas referências são sinalizados todos os autores IPEN, ao clicar nesse símbolo
será aberta uma nova página correspondente à aquele autor – trata-se da página do pesquisador.

O gerenciamento do Repositório está a cargo da Biblioteca do IPEN. Constam neste RI, até o presente momento 20.950 itens que tanto podem ser artigos de periódicos ou de eventos nacionais e internacionais, dissertações e teses, livros, capítulo de livros e relatórios técnicos. Para participar do RI-IPEN é necessário que pelo menos um dos autores tenha vínculo acadêmico ou funcional com o Instituto. Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.
A elaboração do projeto do RI do IPEN foi iniciado em novembro de 2013, colocado em operação interna em julho de 2014 e disponibilizado na Internet em junho de 2015. Utiliza o software livre Dspace, desenvolvido pelo Massachusetts Institute of Technology (MIT). Para descrição dos metadados adota o padrão Dublin Core. É compatível com o Protocolo de Arquivos Abertos (OAI) permitindo interoperabilidade com repositórios de âmbito nacional e internacional.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.
Operando, inicialmente como uma base de dados referencial o Repositório foi disponibilizado na atual plataforma, em junho de 2015. No Repositório está disponível o acesso ao conteúdo digital de artigos de periódicos, eventos, nacionais e internacionais, livros, capítulos, dissertações, teses e relatórios técnicos.
