Abstract:
Acidic environments, such as in inflamed tissues, favor the charged form of local anesthetics (LA). Hence, these drugs show less cell permeation and diminished potency. Since the analgesic capsaicin (CAP) triggers opening of the TRPV1 receptor pore, its combination with LAs could result in better uptake and improved anesthesia. We tested the above hypothesis and report here for the first time the analgesia effect of a two-drug combination (LA and CAP) on an inflamed tissue. First, CAP solubility increased up to 20 times with hydroxypropyl-beta-cyclodextrin (HP-β-CD), as shown by the phase solubility study. The resulting complex (HP-β-CD-CAP) showed 1:1 stoichiometry and high association constant, according to phase-solubility diagrams and isothermal titration calorimetry data. The inclusion complex formation was also confirmed and characterized by differential scanning calorimetry (DSC), X-ray diffraction, and 1H-NMR. The freeze-dried complex showed physicochemical stability for at least 12 months. To test in vivo performance, we used a pain model based on mouse paw edema. Results showed that 2% mepivacaine injection failed to anesthetize mice inflamed paw, but its combination with complexed CAP resulted in pain control up to 45 min. These promising results encourages deeper research of CAP as an adjuvant for anesthesia in inflamed tissues and cyclodextrin as a solubilizing agent for targeting molecules in drug delivery.

Abstract:
The goal of this study was to perform a clinical and molecular investigation in an eight-year-old female child diagnosed with hypophosphatasia (HPP). The proband and her family were evaluated by medical and dental histories, biochemical analyses, radiographic imaging, and genetic analysis of the tissue-nonspecific alkaline phosphatase (ALPL) gene. A bioinformatic analysis was performed to predict the structural and functional impact of the point mutations in the tissue-nonspecific alkaline phosphatase (TNSALP) molecule and to define their potential contribution to the phenotype. We identified a novel combination of heterozygous ALPL missense variants in the proband, p.Ala33Val and p.Asn47His, compatible with an autosomal recessive mode of inheritance and resulting in skeletal and dental phenotypes. Computational modeling showed that the affected Asn47 residue is located in the coil structure close to the N-terminal α-helix, whereas the affected Ala33 residue is localized in the N-terminal α-helix. Both affected residues are located close to the homodimer interface, suggesting they may impair TNSALP dimer formation and stability. Clinical and biochemical follow-up revealed improvements after six years of ERT. Reporting this novel combination of ALPL variants in childhood HPP provides new insights into genotype–phenotype associations for HPP and specific sites within the TNSALP molecule potentially related to a childhood-onset HPP and skeletal and dental manifestations. Beneficial effects of ERT are implicated in skeletal and dental tissues.

Abstract:
The purpose of our work was to select phages displaying peptides capable of binding
to vascular markers present in human atheroma, and validate their capacity to target the vascular
markers in vitro and in low-density lipoprotein receptor knockout (LDLr/) mouse model of
atherosclerosis. By peptide fingerprinting on human atherosclerotic tissues, we selected and isolated
four different peptides sequences, which bind to atherosclerotic lesions and share significant similarity
to known human proteins with prominent roles in atherosclerosis. The CTHRSSVVC-phage peptide
displayed the strongest reactivity with human carotid atherosclerotic lesions (p < 0.05), when
compared to tissues from normal carotid arteries. This peptide sequence shares similarity to a
sequence present in the fifth scavenger receptor cysteine-rich (SRCR) domain of CD163, which
appeared to bind to CD163, and subsequently, was internalized by macrophages. Moreover, the
CTHRSSVVC-phage targets atherosclerotic lesions of a low-density lipoprotein receptor knockout
(LDLr/) mouse model of atherosclerosis in vivo to High-Fat diet group versus Control group. Tetraazacyclododecane-1,4,7,10-tetraacetic acid-CTHRSSVVC peptide (DOTA-CTHRSSVVC) was
synthesized and labeled with 111InCl3 in >95% yield as determined by high performance liquid
chromatography (HPLC), to validate the binding of the peptide in atherosclerotic plaque specimens.
The results supported our hypothesis that CTHRSSVVC peptide has a remarkable sequence for the
development of theranostics approaches in the treatment of atherosclerosis and other diseases.

Abstract:
The mechanisms of action of photobiomodulation (PBM) in oral mucositis (OM) are not
completely elucidated. To enlighten the role of PBM in the evolution of epithelial maturity in OM
ulcers, the present study evaluated the effect of PBM with red ( ) wavelength of 660 nanometers (nm)
and infrared of 780 nm in radio-induced OM wounds on the tongue of rats, eight and twenty days
after irradiation with single dose of 20 Gy. The percentage area corresponding to positive staining
for cytokeratin 10 (CK10) and 14 (CK14) proteins was evaluated in the epithelial area of the lesions,
using an immunohistochemical technique (IHC), 8 and 20 days after the induction of lesions, and
compared with an untreated control group. CK10 was significantly more expressed in the group
treated with 660 nm PBM. CK14 did not show quantitative differences between the groups evaluated.
However, whereas in the groups treated with PBM, CK14 was already restricted to the basal layer
of the epithelium, as expected in healthy epithelia, in control group it was also expressed in upper
layers of the epithelium. In this work, PBM was able to improve epithelial maturity of the repaired
OM wound, especially in the 660 nm group.

Abstract:
Human thyrotropin (hTSH) is a glycoprotein with three potential glycosylation sites: two in
the -subunit and one in the -subunit. These sites are not always occupied and occupancy is
frequently neglected in glycoprotein characterization, even though it is related to folding, trafficking,
initiation of inflammation and host defense, as well as congenital disorders of glycosylation (CDG).
For the first time N-glycoprofiling analysis was applied to the site-occupancy determination of
two native pituitary hTSH, in comparison with three recombinant preparations of hTSH, a widely
used biopharmaceutical. A single methodology provided the: (i) average N-glycan mass; (ii) mass
fraction of each monosaccharide and of sulfate; and (iii) percent carbohydrate. The results indicate
that the occupancy (65%–87%) and carbohydrate mass (12%–19%) can be up to 34%–57% higher in
recombinant hormones. The average glycan mass is 24% lower in pituitary hTSH and contains ~3-fold
fewer moles of galactose (p < 0.005) and sialic acid (p < 0.01). One of the two native preparations,
which had the smallest glycan mass together with the lowest occupancy and GalNAc, sulfate,
Gal and sialic acid contents, also presented the lowest in vivo bioactivity and circulatory half-life.
The methodology described, comparing a recombinant biopharmaceutical to its native equivalent,
can be applied to any physiologically or clinical relevant glycoprotein.

Abstract:
This in vitro study evaluated the compositional, crystalline, and morphological effects promoted by Nd:YAG laser on root dentin, and verified the effects of laser and topical acidulated phosphate fluoride application (APF-gel) on dentin erosion. 180 bovine dentin slabs were randomized into 4 groups (n = 45): G1–untreated, G2–APF-gel (1.23% F−, 4 min), G3–Nd:YAG (1064 nm, 84.9 J/cm2, 10 Hz), and G4–APF-gel application followed by Nd:YAG laser irradiation. The compositional, crystalline, and morphological effects promoted by treatments were investigated on five samples of each experimental group. The other samples were submitted to a 5-day, 10-day, or 15-day erosive and abrasive demineralization and remineralization cycling in order to create erosion lesions. The area and depth of lesions, as well as the optical attenuation coefficient, were assessed, and all data were statistically analysed (p < 0.05). Nd:YAG laser promoted the reduction of carbonate, the formation of tetracalcium phosphate, as well as the melting and recrystallization of the dentin surface. Laser significantly decreased the area and depth of erosion lesions and altered the optical attenuation coefficient when compared to untreated and APF-gel groups, but the association of APF-gel and laser did not promote an additional effect. Nd:YAG laser irradiation can be a promissory treatment to prevent dentin erosion and the abrasion process.

Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). A elaboração do projeto do RI do IPEN foi iniciado em novembro de 2013, colocado em operação interna em julho de 2014 e disponibilizado na Internet em junho de 2015. Utiliza o software livre Dspace, desenvolvido pelo Massachusetts Institute of Technology (MIT). Para descrição dos metadados adota o padrão Dublin Core. É compatível com o Protocolo de Arquivos Abertos (OAI) permitindo interoperabilidade com repositórios de âmbito nacional e internacional.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.
Operando, inicialmente como uma base de dados referencial o Repositório foi disponibilizado na atual plataforma, em junho de 2015. No Repositório está disponível o acesso ao conteúdo digital de artigos de periódicos, eventos, nacionais e internacionais, livros, capítulos, dissertações, teses e relatórios técnicos.

O gerenciamento do Repositório está a cargo da Biblioteca do IPEN. Constam neste RI, até o presente momento 20.950 itens que tanto podem ser artigos de periódicos ou de eventos nacionais e internacionais, dissertações e teses, livros, capítulo de livros e relatórios técnicos. Para participar do RI-IPEN é necessário que pelo menos um dos autores tenha vínculo acadêmico ou funcional com o Instituto. Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN.
