Could light-based technologies improve stem cell therapy for skin wounds?: A systematic review and meta-analysis of preclinical studies.

Abstract:
Several diseases or conditions cause dermatological disorders that hinder the process of skin repair. The search for novel technologies has inspired the combination of stem cell (SC) and light-based therapies to ameliorate skin wound repair. Herein, we systematically revised the impact of photobiomodulation therapy (PBM) combined with SCs in animal models of skin wounds and quantitatively evaluated this effect through a meta-analysis. For inclusion, SCs should be irradiated in vitro or in vivo, before or after being implanted in animals, respectively. The search resulted in nine eligible articles, which were assessed for risk of bias. For the meta-analysis, studies were included only when PBM was applied in vivo, five regarding wound closure, and three to wound strength. Overall, a positive influence of SC + PBM on wound closure (mean difference: 9.69; 95% CI: 5.78–13.61, P < 0.00001) and strength (standardized mean difference: 1.7, 95% CI: 0.68–2.72, P = 0.001) was detected, although studies have shown moderate to high heterogeneity and a lack of information regarding some bias domains. Altogether, PBM seems to be an enabling technology able to be applied postimplantation of SCs for cutaneous regeneration. Our findings may guide future laboratory and clinical studies in hopes of offering wound care patients a better quality of life.

Abstract:
Leishmanioses são doenças parasitárias desenvolvidas por protozoários do gênero Leishmania. A forma cutânea abrange lesões destrutivas e ulceradas com diversas limitações no tratamento. Terapias alternativas são fundamentais devido à grande relevância da doença e elevada incidência. Nesse contexto, a terapia fotodinâmica (PDT) tem sido introduzida devido ao baixo custo, baixa toxicidade, praticidade e sem relatos de resistência na literatura. Neste trabalho, dezesseis camundongos BALB/c foram infectados com 1.106 parasitos de Leishmania(L) amazonensisno membro posterior esquerdo e acompanhados por 4 semanas até o surgimento da lesão. Após esse período, os animais foram submetidos à PDT usando um diodo emissor de luz (λ=660 ±22 nm) e azul de metileno (100 μM), com fluências de 50, 100 e 150J/cm² e acompanhados durante 3 semanas. Animais controle não receberam tratamento. O tamanho da lesão e escala de dor foram mensurados utilizando um paquímetro e filamentos von Frey, respectivamente. A quantificação da carga parasitária foi realizada através do método de diluição limitante. Os resultados demonstraram que, nas semanas 2 e 3 após tratamento, ocorreu diminuição da lesão e alívio de dor nos animais que receberam 150J/cm², sugerindo que a PDT promoveu melhora clínica através da modulação do processo inflamatório.

Abstract:
INTRODUÇÃO. Os avanços na área de tecnologias baseadas em luz vêm
contribuindo fortemente para o desenvolvimento da medicina regenerativa.
Assim, a aplicação da luz de baixa irradiância (LBI) em diferentes regimes de
irradiação poderia estimular a proliferação e acelerar a diferenciação de
células-tronco mesenquimais (CTMs) em tenócitos, que poderiam ser utilizados
na construção de um tendão artificial para reparação de lesões tendíneas.
OBJETIVO: Avaliar os efeitos de diferentes comprimentos de onda na
proliferação e diferenciação de células mesenquimais em tenócitos.
METODOLOGIA: A confirmação da pluripotência das células foi realizada
através de citometria de fluxo. Células-tronco humanas derivadas de tecido
adiposo (Parecer CEP: 3.596.852) foram isoladas e colocadas em meio de
cultura contendo ou não fatores de diferenciação para tenócitos. As células
foram irradiadas a cada 48 h, durante 9 dias, com diferentes diodos emissores
de luz (LEDs): azul (λ=450 ± 10 nm), verde (λ=520 ± 10 nm) e vermelha
(λ=660 ± 10 nm), com irradiância de 23,3 mW/cm2 durante 17 min e 10 s,
entregando uma densidade de energia de 24 J/cm2 por sessão. O grupo
controle não recebeu tratamento. Após o quinto e o nono dia, foi verificada a
proliferação celular por cristal violeta e ensaio de MTT, bem como a
diferenciação em tenócitos por microscopia de fluorescência. RESULTADOS:
Observou-se um aumento na proliferação celular quando usado o LED de
emissão vermelha. Os LEDs de emissão azul e verde mostraram viabilidade
celular semelhante ou abaixo do grupo controle. Por outro lado, embora todos
os comprimentos de onda tenham promovido marcação positiva para colágeno
Tipo I e colágeno tipo III, o LED de emissão vermelha apresentou maior
densidade celular. CONCLUSÃO: Nossos resultados indicam que o LED de
emissão vermelha, nas condições deste estudo, é capaz de aumentar a
proliferação celular e contribuir para melhor diferenciação de CTMs em
tenócitos.

Abstract:
Impact of low-level laser irradiation on tumor cell lines remains
controversial. Vascular endothelial growth factor (VEGF) is a key
molecule to form new blood vessels, which contribute for cancer
development and growth. The aim of this study was to evaluate
the effects of different light fluences on human melanoma
SKMEL 37 cells and murine melanoma B16F10 cells using a
near infrared laser (λ= 780 nm) with output power of 40 mW
delivering energies of 1.2, 3.6 and 6 J (fluences of 30, 90 and
150 J/cm2, respectively). The cell lines were irradiated 24 h after
they were seeded in a 96-well plate at a density of 5x103 cells
per well, in triplicate at three different days. Following irradiation,
both cell line supernatants were stored in Eppendorf tubes at -
20°C until VEGF-A expression measurement. Specific ELISA kits
were used according to cell line (murine or human). Samples and
standard solutions were added in a 96-well plate antibody-coated
and incuba ted over night at 4°C. Reagent dilution and set time
followed fabricant instructions. The stop solution was added and
the absorbance was read in a microplate reader at 450 nm.
Results showed a non-statistically significant difference among
treated and control groups for both cell lines. These findings
indicate that irradiation with near infrared laser does not influence
VEGF expression on melanoma cell lines regardless the fluence
used and should be tested to prevent cancer growth in preclinical
assays.

Abstract:
INTRODUÇÃO. A Fotobiomodulação (FBM) é uma modalidade terapêutica que
apresenta ampla gama de aplicações clínicas e seus mecanismos de atuação
na inflamação ainda são objeto de investigação em diferentes tipos de modelos
experimentais. O zebrafish é um modelo animal amplamente utilizado pela
indústria farmacêutica para investigação da resposta inflamatória frente a
diferentes fármacos. OBJETIVO: O objetivo deste estudo é estabelecer um
modelo de estudo da FBM em zebrafish após indução de processo
inflamatório. METODOLOGIA: Após aprovação pelo comite de ética (CEUA
336/2017) os animais foram divididos em grupos a saber G1 sem intervenção,
G2 Injeção de solução salina na cavidade celomática, G3 injeção de 3,5% de
solução de carragenina na cavidade celomática, G4, G5 e G6 injeção de 3,5%
de solução de carragenina na cavidade celomática e irradiação com laser de
λ=660nm, com 100mW entregando 3J, 4J e 20J de energia respectivamente.
Imagens de tomografia por coerência óptica (OCT) e histologia foram
realizados em diferentes momentos para avaliação dos efeitos.
RESULTADOS: Os resultados observados por OCT apresentam evolução do
quadro inflamatório após irradiação, confrontando os dados da OCT com os
achados histológicos a energia de 20J apresentou o melhor resultado na
resolução do edema e da resposta inflamatória promovida pela carragenina.
CONCLUSÃO: O zebrafish pode ser um novo modelo de estudo para
investigação dos efeitos da FBM e devido a suas características a adequação
das dosimetrias para este modelo animal merecem mais investigação.

Abstract:
A síndrome metabólica (SM) é uma condição clínica que agrupa uma variedade de morbidades, como intolerância à glicose e obesidade. Na obesidade, o tecido adiposo branco (TAB) apresenta características inflamatórias que interferem na ação da insulina, levando à ocorrência de Diabetes do tipo 2. O tecido adiposo marrom (TAM), que tem como principal função a termogênese através da oxidação mitocondrial de cadeias carbônicas, se encontra hiporresponsivo aos estímulos clássicos na SM, como, por exemplo, a exposição ao frio. Estratégias para modular os processos inflamatórios do TAB e ativar o metabolismo do TAM podem atenuar as consequências da SM. Os reconhecidos efeitos anti-inflamatórios e de ativação do metabolismo mitocondrial da fotobiomodulação (PBM) indicam que essa poderia ser uma proposta terapêutica para a SM. Sendo esse o nosso foco de estudo, camundongos adultos, machos, da linhagem C57BL/6 receberam dieta hiperlipídica para indução da SM. Os animais foram então irradiados usando um dispositivo LED sobre a superfície abdominal (λ = 850 nm) ou interescapular (λ = 660 nm) para modular a inflamação do TAB ou ativar o TAM, respectivamente. O tratamento consistiu em 6 sessões de irradiação, distribuídas no decorrer de 21 dias. Apesar de não terem apresentado alterações na massa corporal e Índice de Lee, os animais irradiados na região abdominal (HFABD850) apresentaram 50 % menos células inflamatórias no TAB epididimal e também apresentaram melhora no teste de tolerância à glicose 24 h após a última sessão de tratamento. Nos animais obesos irradiados na região interescapular (HFTAM660), as irradiações promoveram aumento de duas vezes na massa do TAM, além de aumento da temperatura dorsal e da captação de 18F-FDG após exposição a baixas temperaturas. O soro desses animais (HFTAM660) também se mostrou mais semelhante ao de animais eutróficos. Nossos achados indicam que a PBM, nos parâmetros investigados, pode ser aplicada ao tratamento da SM.

Abstract:
Photobiomodulation (PBM) has been gaining importance in a wide range of medical fields in the past few
years, particularly in stem cell-based regenerative medicine. Improving in vitro cell proliferation,
differentiation and viability are ways where PBM could play a pivotal role optimizing biotechnological and
bioengineering applications. Here we investigated whether different wavelengths (blue, green and red)
would promote distinct outcomes in human adipose-derived stem cells (hADSCs) cultured in regular and
supplemented media for tenocyte differentiation. MATERIALS AND METHODS. Freshly isolated hADSCs
were cultured in a specific stem cell medium (MSCGM, Lonza), DMEM or a tenogenic medium (TEN-M:
DMEM supplemented with growth factors and ascorbic acid). Cells were irradiated every 48 h (23.28
mW/cm 2, 17 min 10 s delivering 24 J/cm 2 per session) using a LED irradiator (LEDbox, BioLambda).
MTT and crystal violet assays were used to evaluate cell metabolic activity and proliferation. Red
wavelength (660 nm) significantly increased metabolic activity after five irradiations, but only for cells
cultured in TEN-M. Oppositely, blue (450 nm) and green (520 nm) light decreased both cell proliferation
and metabolic rate, with more pronounced effects for blue light in TEN-M. Considering these findings, we
examined whether irradiating only the media would generate toxic compounds that could impair cell
viability. We therefore assessed reactive oxygen species (ROS) production by p-nitrosodimethylaniline/
histidine assay while irradiating the three different media under the same conditions
as mentioned above. Immediately after blue and green light exposure, an increment in ROS production
was observed for DMEM and TEN-M, that continuously increased until reaching between 4.5 and 7.1 μM
one-hour after irradiation – with higher values for TEN-M exposed to blue light. Since no significant ROS
formation was observed following red light exposure, we concluded that medium composition was
responsible for the different effects on metabolic activity and proliferation observed after irradiation with
different wavelengths.

Abstract:
INTRODUCTION
Photobiomodulation (PBM) has been gaining importance in
a wide range of medical fields in the past few years, particularly
in stem cell-based regenerative medicine. Improving
in vitro cell proliferation, differentiation and viability are
ways where PBM could play a pivotal role optimizing biotechnological
and bioengineering applications.
OBJECTIVES
Here we investigated whether different wavelengths (blue,
green and red) would promote distinct outcomes in human
adipose-derived stem cells (hADSCs) cultured in regular and
supplemented media for tenocyte differentiation.
MATERIALS AND METHODS
Freshly isolated hADSCs were cultured in a specific stem cell
medium (MSCGM, Lonza), DMEM or a tenogenic medium
(TEN-M: DMEM supplemented with growth factors and ascorbic
acid). Cells were irradiated every 48 h (23.28 mW/cm 2 ,
17 min 10 s delivering 24 J/cm 2 per session) using a LED irradiator
(LEDbox, BioLambda). MTT and crystal violet assays
were used to evaluate cell metabolic activity and proliferation.
DISCUSSION AND RESULTS
Red wavelength (660 nm) significantly increased metabolic
activity after five irradiations, but only for cells cultured in
TEN-M. Oppositely, blue (450 nm) and green (520 nm) light
decreased both cell proliferation and metabolic rate, with more
pronounced effects for blue light in TEN-M. Considering these
findings, we examined whether irradiating only the media would generate toxic compounds that could impair cell viability. We
therefore assessed reactive oxygen species (ROS) production
by p-nitrosodimethylaniline/histidine assay while irradiating
the three different media under the same conditions as mentioned
above. Immediately after blue and green light exposure,
an increment in ROS production was observed for DMEM and
TEN-M, that continuously increased until reaching between 4.5
and 7.1 μM one-hour after irradiation – with higher values for
TEN-M exposed to blue light.
CONCLUSION
Since no significant ROS formation was observed following
red light exposure, we concluded that medium composition
was responsible for the different effects on metabolic activity
and proliferation observed after irradiation with different
wavelengths.

Abstract:
A síndrome metabólica (SM) é uma condição clínica que agrupa uma variedade de morbidades, como hiperglicemia, pressão arterial elevada, dislipidemia aterogênica e obesidade (particularmente na região abdominal). Nessa conjuntura, os principais tecidos-alvo da ação da insulina sofrem alterações metabólicas que aumentam o risco de ocorrência de doenças cardiovasculares e diabetes tipo 2. As alterações teciduais observadas são caracterizadas por infiltrados de células do sistema imune, especialmente macrófagos. Citocinas pró-inflamatórias, como TNF-α, são liberadas e alcançam a corrente sanguínea, promovendo nesses indivíduos um estado de inflamação crônica e sistêmica. O tecido adiposo intra-abdominal parece ser de particular importância no estabelecimento desse quadro inflamatório, e estratégias direcionadas no sentido de modular os processos inflamatórios nesse tecido podem atenuar as consequências da SM. Os reconhecidos benefícios da terapia com luz de baixa potência em condições inflamatórias nos permitem supor que essa poderia ser uma proposta terapêutica para a SM. Sendo esse o nosso foco de estudo, camundongos adultos, machos, das linhagens C57BL/6 e BALB/c receberam dieta hiperlipídica durante 8 semanas para indução do quadro de SM. Os animais foram então irradiados sobre a superfície abdominal no decorrer de 21 dias, usando um LED (λ = 850 nm, 6 sessões, 300 s por sessão, potência = 60 mW, fluência = 6 J/cm², taxa de fluência = 19 mW/cm²). Antes e durante o tratamento, amostras de sague foram coletadas para quantificação de glicose, colesterol total e triglicérides plasmáticos. Considerando os parâmetros de irradiação adotados, a terapia com luz de baixa potência não se mostrou efetiva para alterar massa corporal, glicemia, colesterol total e triglicérides de camundongos alimentados com dieta hiperlipídica.

Abstract:
Currently, antimicrobial photodynamic therapy (APDT) is limited to the local treatment of topical infections, and a platform that can deliver the photosensitizer to internal organs is highly desirable for non-local ones; SPIONs can be promising vehicles for the photosensitizer. This work reports an innovative application of methylene blue (MB)-superparamagnetic iron oxide nanoparticles (SPIONs). We report on the preparation, characterization, and application of MB-SPIONs for antimicrobial photodynamic therapy. When exposed to light, the MB photosensitizer generates reactive oxygen species (ROS), which cause irreversible damage in microbial cells. We prepare SPIONs by the co-precipitation method. We cover the nanoparticles with a double silica layer – tetraethyl orthosilicate and sodium silicate – leading to the hybrid material magnetite-silica-MB. We characterize the as-prepared SPIONs by Fourier transform infrared spectroscopy, powder X-ray diffraction, and magnetic measurements. We confirm the formation of magnetite using powder X-ray diffraction data. We use the Rietveld method to calculate the average crystallite size of magnetite as being 14 nm. Infrared spectra show characteristic bands of iron‑oxygen as well as others associated with silicate groups. At room temperature, the nanocomposites present magnetic behavior due to the magnetite core. Besides, magnetite-silica-MB can promote ROS formation. Thus, we evaluate the photodynamic activity of Fe3O4-silica-MB on Escherichia coli. Our results show the bacteria are completely eradicated following photodynamic treatment depending on the MB release time from SPIONs and energy dose. These findings encourage us to explore the use of magnetite-silica-MB to fight internal infections in preclinical assays.

Abstract:
Background: Artificial insemination is widely employed in poultry, but high degrees of bacterial contamination are often observed in semen because of its passage through the cloaca. Consequently, most semen extenders for birds have antibiotics that could aggravate bacterial resistance. Methods: We evaluated the potential of antimicrobial photodynamic therapy (PDT) as an alternative to the use of antibiotics, and assessed whether changes in concentration and incubation time with methylene blue (MB), radiant exposure, and irradiance of light affect spermatozoa activity and bacteria in chicken semen. Results: Incubation with MB (< 25 µM) did not alter sperm motility, regardless of the pre-irradiation time (PIT, 1 or 5 min). Following 1 min of PIT with MB at 10 µM, samples were irradiated for 30, 60, 120, and 180 s at irradiances of 44, 29, and 17 mW/ cm² (660 nm LedBox). MB and light alone did not interfere with the analyzed parameters. However, when both factors were associated, increases in light dose led to greater reductions in sperm parameters, regardless of the irradiance used. Besides, PDT conditions that were less harmful to spermatozoa were not able to significantly reduce bacterial colonies in chicken semen. Conclusions: A failure in MB selectivity could explain unsuccessful bacterial reduction following PDT. Further research involving other photosensitizers or conjugating molecules to MB to target microbial cells is needed for PDT application in poultry breeders.

Abstract:
Gold nanoshells (silica core/gold shell) have fascinating
optical extinction spectrum within visible to near infrared
range. In this work, we investigate the optical properties of
Au nanoshells and evaluate the feasibility of their use on
photodynamic therapy. Three dimensional finite element
simulation and experimental analyses were explored on
the assessment of the localized surface plasmon resonance
spectrum and spatial distribution of the electromagnetic
field enhancement near metallic nanoshells. In addition,
the interaction of Au nanoshells with methylene blue
(MB) photosensitizer was appraised, and 3.2—fold metal
enhanced single oxygen generation was observed. Lastly,
we investigate the effects of MB-mediated photoinactivation
on Staphylococcus aureus assisted by gold nanoshells.
The use of nanoshells on the photoinactivation
procedure reduced 3 the required illumination time for
total eradication of bacterial cells. Our results indicate that
Au nanoshells are promising candidates to enhance the
photodynamic effect on bacterial cells.

Abstract:
The increasing resistance to antimicrobials worldwide requires new strategies to fight infectious diseases. Antimicrobial photodynamic therapy (APDT) emerges in this scenario as a promising approach. APDT is a light-based therapy combined to a photosensitizer drug and oxygen that kills microorganisms via oxidative stress. Due to its mechanism of action, i.e., generalized oxidation of all cell structures, bacterial resistance to APDT is very improbable. Currently, however, APDT is limited to local treatments. Thus, a platform to deliver the photosensitizer drug in internal organs is urgently necessary. Superparamagnetic iron oxide nanoparticles (SPIONs) can be easily directed to target sites using an external magnetic field, which made them excellent drug delivery systems. In addition, their large surface to volume ratio allows the functionalization of therapeutic molecules on their surface. In this work, we synthesized and characterized SPIONs covered by methylene blue (MB)-entrapped silica (SPIONs-silica-MB) and investigated their potential combined to APDT to kill Escherichia coli, the most common bacteria found in urinary tract infections. We synthesized SPIONs by the co-precipitation of iron (II) and (III) chlorides in the presence of a weak base and covered them with a double layer of silica leading to the hybrid material magnetite-silica-MB. SPIONS were characterized by Fourier transform infrared spectroscopy, powder X-ray diffraction, and magnetic measurements. Kinetics of MB release and production of singlet oxygen from SPION-silica-MB were also obtained. The average crystallite size of magnetite was found to be 14 nm. Infrared spectra showed characteristic bands of iron-oxygen and others associated with silicate groups. At room temperature, the nanoplatform presented magnetic behavior due to the magnetite core. MB release increased exponentially until 18 h, when it reached a plateau. Singlet oxygen was spontaneously released from SPIONs-silica-MB. To verify the photodynamic activity of SPIONs-silica-MB on bacterial cells, 1x10^7 colony forming units of E. coli were suspended in SPIONs-silica-MB PBS solution (50 microM of MB) and were irradiated using a red LED (625 ± 20 nm) of 235 mW over an orbital shaker to prevent precipitation during 5, 10, 15 and 20 min at three different MB release times: 6, 12 and 22 h. Proper controls were established and showed no killing. In contrast, SPIONs-silica-MB-mediated APDT promoted bacteria inactivation depending on both the time of MB release and irradiation time. Our results show that bacteria are completely eradicated following 22 h of MB release and 20 min of irradiation. These findings motivate the use of SPIONs-silica-MB to mediate APDT against nonlocal infectious diseases.

Abstract:
Mammal fat consists ofwhite and brown adipose tissues (WATand BAT).
Although most fat is energy-storing WAT, brown adipocytes dissipate
energy as heat. Obese subjects exhibit BAT hyporesponsiveness to classical
stimuli as cold, hence increasing BAT thermogenic capacity
could be an i n t er e s t i n g t a rget f o r t r eating obesity.
Photobiomodulation (PBM) seems to be a feasible alternative to
activate BAT, although it has not been applied for this purpose
until now. In this study, adult C57Bl/6 mice were fed with a
hyperlipidic diet (HF) during 15 weeks to induce obesity. Then,
they were divided into two groups: HF and HF660. The
interscapular BAT of HF660 mice was exposed to a single irradiation
using a 662 ± 20 nm LED (5.7 Jcm-2 at 19 mWcm-2,
delivered during 300 s). The HF group was sham-irradiated.
Eutrophic animals were also used. Animals received 200 mCi of
18F-FDG and were exposed to cold for basal register of PET-CT
images. After PBM or sh am treat ment, mice were once again
exposed to cold before PET-CT image acquisition. Our results
showed that PBM was able to activate BAT in obese mice
through higher glucose uptake. In contrast, PBM and shamirradiated
normal fed mice presented similar levels of glucose
uptake. Taking together, our findings suggest that PBM applied
to the BAT is able to promote glucose uptake by obese mice and
could be an interesting approach to manage obesity and its correlated
morbidities.

Abstract:
Ventilator-associated pneumonia (VAP) is an infection that
arises after endotracheal intubation affecting patients under
intensive care. The presence of the endotracheal tube
(ETT) is a risk factor since it is colonized by multispecies
biofilm. Antimicrobial photodynamic therapy (aPDT) could
be a strategy to decontaminate ETTs. We verify if methylene
blue (MB) associated with external illumination of the
ETT could be an alternative to destroy biofilm. We performed
an in vitro and ex vivo study. In vitro study was
performed with P. aeruginosa biofilm grew over ETT for
7 days. After treatment, the surviving cells were cultured
for 3 days and the biofilm was analyzed by crystal violet
absorbance. Ex vivo study employed ETT obtained from
extubated patients. aPDT was performed with MB (100 μM)
and red LED (k = 640±20 nm). We quantified the biofilm
thickness and used scanning electron microscopy and fluorescence
technique to verify morphological and functional
changes after aPDT. Our results showed that bacteria
remain susceptible to aPDT after sequential treatments. We
also attested that aPDT can reduce biofilm thickness,
disrupt biofilm attachment from ETT surface and kill
microbial cells. These data suggest that aPDT should
be investigated to decrease VAP incidence via ETT
decontamination.

Abstract:
Cutaneous leishmaniasis is a neglected parasitic disease that leads to destructive lesions. The emergence of drug resistance has been a global concern over the past years. Photodynamic therapy (PDT) mediated by a red LED and methylene blue (MB) involves the overproduction of oxidative stress, which oxidizes several cellular biomolecules and prevents the selection of resistant strains. Herein, we investigated the potential of PDT mediated by MB against wild-type and miltefosine-resistant strains of Leishmania amazonensis. As a result, both strains were susceptible to PDT, thus encouraging us to seek the best conditions to overcome the drug resistance problem in cutaneous leishmaniasis.

Abstract:
Regulation of inflammation is a critical process for maintaining physiological homeostasis. The λ-carrageenan (λ-CGN) is a mucopolysaccharide extracted from the cell wall of red algae (Chondrus crispus) capable of inducing acute intestinal inflammation, which is translated into the production of acute phase reactants secreted into the blood circulation. However, the associated mechanisms in vertebrates are not well understood. Here, we investigated the crucial factors behind the inflammatory milieu of λ-CGN-mediated inflammation administered at 0, 1.75, and 3.5% (v/w) by i.p. injection into the peritoneal cavity of adult zebrafish (ZF) (Danio rerio). We found that polymorphonuclear leukocytes (neutrophils) and lymphocytes infiltrating the ZF peritoneal cavity had short-term persistence. Nevertheless, they generate a strong pattern of inflammation that affects systemically and is enough to produce edema in the cavity. Consistent with these findings, cell infiltration, which causes notable tissue changes, resulted in the overexpression of several acute inflammatory markers at the protein level. Using reversed-phase high-performance liquid chromatography followed by a hybrid linear ion-trap mass spectrometry shotgun proteomic approach, we identified 2938 plasma proteins among the animals injected with PBS and 3.5% λ-CGN. First, the bioinformatic analysis revealed the composition of the plasma proteome. Interestingly, 72 commonly expressed proteins were recorded among the treated and control groups, but, surprisingly, 2830 novel proteins were differentially expressed exclusively in the λ-CGN-induced group. Furthermore, from the commonly expressed proteins, compared to the control group 62 proteins got a significant (p < 0.05) upregulation in the λ-CGN-treated group, while the remaining ten proteins were downregulated. Next, we obtained the major protein-protein interaction networks between hub protein clusters in the blood plasma of the λ-CGN induced group. Moreover, to understand the molecular underpinnings of these effects based on the unveiled protein sets, we performed a bioinformatic structural similarity analysis and generated overlapping 3D reconstructions between ZF and humans during acute inflammation. Biological pathway analysis pointed to the activation and abundance of diverse classical immune and acute phase reactants, several catalytic enzymes, and varied proteins supporting the immune response. Together, this information can be used for testing and finding novel pharmacological targets to treat human intestinal inflammatory diseases.

Abstract:
The present study evaluated the effectiveness of extracts of commercially available Curcuma longa, Citrus lemon,
Hamamelis virginiana and Hypericum perforatum as photosensitizers in Antimicrobial Photodynamic Therapy
(aPDT). Each photosensitizer (PS) was analyzed in a spectrophotometer between 350 and 750 nm to determine
the ideal light source. Once the absorption bands were determined, three light sources were selected. To determine
the concentration of use, the compounds were tested at different concentrations on bovine dentin
samples to evaluate the risk of staining. Once the concentration was determined, the PSs were evaluated for dark
toxicity and phototoxicity on fibroblast and bacteria culture. Each compound was then irradiated with each light
source and evaluated for the production of reactive oxygen species (ROS). The bacterial reduction was tested on
E. faecalis culture in planktonic form and on biofilm using an energy of 10 J and an Energy Density of 26 J/cm2.
The tested compounds exhibited light absorption in three bands of the visible spectrum: violet (405 nm), blue
(460 nm) and red (660 nm). At a 1:6 concentration, none of the compounds caused tooth staining as they did not
exhibit significant toxicity in the cells or bacterial suspension. Additionally, significant ROS production was
observed when the compounds were irradiated at each wavelength. When aPDT was performed on the plactonic
and biofilm bacteria, significant microbial reduction was observed in both cases, reaching a reduction of up to
5Logs. In conclusion, extracts of Curcuma longa, Citrus lemon, Hamamelis virginiana and Hypericum perforatum
exhibited potential for use as photosensitizing agents in aPDT.

Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.
