Abstract:
A papaína, enzima proteolítica extraída do látex do fruto verde do mamão papaia
intermolecular e esterilização do meio, simultaneamente.

Abstract:
The recent concern related to skin compromised patients, apart from the nature of the
condition itself, such as wounds, chronic ulcers, or burns among others, has triggered
and highlighted the importance of the development of artificial skins available in allogeneic
donor tissue banks and/or scaffolds, composed by a wide variety of biocompatible,
biodegradable and bioactive biomaterials. Within this context, tissue engineering has been
in expansion as an attempt to overcome difficulties faced in such situations. The application
of scaffolds, produced or not by nanotechnology, in the skin of a patient induces cells to
proliferate and get organized on extracellular matrix regenerating tissue. Ionizing radiation
is a particularly useful technology capable of promoting sterilization and cross-linking of
the scaffold structure thus offering several possibilities for the development of advanced
systems suitable for cell growth. Taking into account the variety of clinical applications of
tissue engineering, the aim of this study was to investigate by means of histological tests,
chemistries and non-destructive tests, the interaction of mesenchymal stem cells grown
in vitro in conjunction with different frameworks in order to understand how the mesenchymal
stem cells behave in different niches. Among those collagen, PVA, chitosan, PDLLA
scaffolds were the systems of choice and
-irradiation was applied for sterilization of the
systems, as well as cross-linking for the PVA based scaffold. Thus this work allowed the
achievement of dermo-epidermic matrices populated by epidermal cells that make up the
skin and will contribute to the development of a more robust and useful material to be used
in several treatments.

Abstract:
Lignin is a carbon renewable source and has been widely explored in different fields in
the last years, especially in biomaterials as dressing and other biomedical devices due
its natural origin and low cost1. Poly(ethylene oxide) (PEO) is a synthetic polymer
largely used in biomedical applications due its important characteristics such as high
hydrophilicity, non-toxicity and ease of process2. The present work aimed to develop
a lignin-PEO based dressing for wound treatment by casting. In specific terms, three
different polymer blends were formulated using a range of 3 to 10% (w/v) lignin was
tested with the addition of 1 to 3% PEO. Lignin was solubilized in aqueous solution
(pH>13) alkalized with sodium hydroxide and PEO was solubilized in distilled water.
The solutions were heated up to 70 ºC and homogenized until complete polymer
dissolution. Then, PEO solution and poly(ethylene glycol) diacrylate (PEGDA) (0.5 -
1.0%) were added to the lignin solution and the blend was mixed for 30 minutes at 70
ºC. Posteriorly the blends were submitted to casting and drying under different
conditions, room temperature for 48 hours and incubated at 40 ºC for 24 hours. A
control sample of 6% lignin was prepared in the same conditions. Samples were
evaluated by physico-chemical and morphological characterizations. The swelling and
gel fraction profiles were assessed as well as thermal behavior by differential scanning
calorimetry. Chemical modifications were evaluated by infrared spectroscopy.
Samples with higher PEGDA content presented minor swelling index. The blends
formulated presented different thermal behavior in comparison with the control.
Infrared spectroscopy pointed some chemical modifications promoted by the
crosslinking agent. In general terms, the material developed presented a potential to
continue been explored as dressing for wound treatment

Abstract:
Bladder cancer (BC) is one of the main diseases that attack the urinary tract and is
globally responsible for 165,000 deaths per year. Bladder tumor may be classified as
non-muscle invasive (superficial) or muscle invasive. About 70% of patients present
the superficial bladder cancer which is treated by transurethral resection for tumor
removal followed by intravesical chemo or immunotherapy. The main challenge
reported in the instillation of chemotherapy is the limited drug residence time in the
bladder as a consequence of urine levels that leads to fast drug removal from the
bladder.1 Hydrogels are chemically or physically crosslinked polymer systems that
form three-dimensional networks with high water absorption capacity.
Carboxymethylcellulose (CMC) is one of the major soluble derivatives of cellulose
widely applied in the medical and pharmaceutical fields due to its biocompatibility,
nontoxicity, biodegradability and film forming ability.2 Polyvinyl Alcohol (PVA) is a
synthetic polymer with wide pharmaceutical and biomedical applications as it is nontoxic,
non-carcinogenic, bioadhesive and easy to process.3 In this work, we developed
a mucoadhesive hydrogel from a CMC and PVA polymer blend for chemotherapeutic
loading and suitable rheological properties for intravesical instillation, especially
designed for treating superficial BC with increased residence time of chemotherapeutic
agent. Three formulations (C1, C2 and C3) at different CMC concentrations were
prepared - 1%, 2% and 3% (w/v), respectively. PVA concentration corresponded to 1%
(w/v) for all formulations. The polymers were separately solubilized in Milli-Q water and
then mixed prior to the addition of 20% (v/v) glycerin to increase the
mucoadhesiveness of the material. The hydrogels were characterized according to
their organoleptic and rheological properties to evaluate the behavior of the material
under tension and temperature. Accelerated stability tests of the pharmaceutical form
were also performed. Comparatively, formulations with glycerin presented improved
mucoadhesiveness, and formulations with 1 and 2% of CMC presented more adequate
rheological behavior for the proposed application. In conclusions, the systems
presented adequate properties for the delivery of chemotherapeutic agents for
optimized BC treatments.

Abstract:
o
grupo com 2% apresentou 0.150 g.L1
e o grupo com 10% apresentou 0.151 g.L1.
Observouse

Abstract:
The oral route is one of the main routes for administration of drugs, however, the gastrointestinal tract is a hostile environment due to pH variation, presence of several digestive enzymes and the intestinal barrier that undermines the permeation of drugs. The aim of this study was to evaluate the ability of papain complexed with β-cyclodextrin to enhancer the permeation of furosemide, as a model drug, in a triple co-culture of Caco-2, HT29-MTX and Raji cells. Papain and the papain-β-cyclodextrin complex were evaluated at 0.3, 0.7 and 1.0 μM and the biophysical integrity of the cell layer was evaluated by RET (Transepitelial Electrical Resistance) at 0, 4, 24, 48 and 72 hours. The epithelium was also stained using DAPI and Alexa Fluor™ 488 Phalloidin. HPLC was employed to quantification of furosemide. The RET results at initial time for all the samples and control were in a range of 267.63 to 318.28 Ω*cm2 and after 72 h this values were raised to a range of 365.14 to 492.64 Ω*cm2. There was a decrease of RET after samples’ application, nevertheless, the results showed that the epithelium presents a recovery, proportional to the time of cell replication, and that this recovery occurs in all samples tested with no significant statistical difference. The RET recovery implies that papain, complexed or not, was not able to kill the cells, corroborating the hypothesis that the action mechanism is the disruption of the tight junction. In addition, the triple co-culture presents a higher resistance to papain action, in comparison with the Caco-2 monolayer assays, emphasizing the importance of testing new drugs, potential candidates for oral formulations, in epitheliums that faithfully mimics what actually happens in in vivo systems. The fluorescent microscopy observation of the cells stained with DAPI and the junctions stained with Alexa Fluor™ 488 Phalloidin, showed that co-culture exhibits microvilli inherent to the intestinal tissue. The results obtained in the triple co-culture model bi-directional transport experiments confirmed the significant increase in furosemide transport indicating the importance of the paracellular route. In conclusion, the triple co-culture model was successfully standardized and papain complexed with β-cyclodextrin acts probably over the tight junctions enhancing the permeation of furosemide.

Abstract:
A síntese será realizada na presença (20%) e ausência do etanol, tampão fosfato 50 Mm pH 7,4 e radiação ionizante a 10 kGy durante 2 horas (5 kGy/hora).

Abstract:
The highlighted role of protein and peptide based delivery systems relies upon the possibility
to develop biocompatible drug carriers featuring site specific delivery, biological affinity
among unique advantages. Recently, a technique for protein nanostructuring by the use
of radiation has been recently reported by our group. Advantages of the use of radiation
over conventional methods are related to the possibility to achieve protein cross-linking
and sterilization in a single step, as well as the capacity to allow the design of nanocarriers
without the need of monomers or toxic cross-linkers. This work reports the use of high
energy irradiation towards the design of size-controlled protein-based nanocarriers and
bioactive nanoparticles, using bovine serum albumin (BSA) and papain as model protein and
protease, respectively, including the state of the art and current advances of the technology.
The technique implies on protein desolvation/solvation techniques followed by cross-linking
by EB radiation or
-irradiation alone, although nanoparticles were also achieved in absence
of the cosolvents. Size-controlled BSA nanocarriers were manufactured up to 80 nm and
papain bioactive nanoparticles up to 12 nm, as determined by dynamic light scattering.
Nanocarrier morphology was evaluated by and negative staining transmission electron
microscopy. Protein cross-linking and changes in aromatic the amino acids were evaluated
by fluorescence measurements. Biocompatibility experimentswere also performed by means
of cytotoxicity and cytokines production. The potential of the systems for the delivery of
radiopharmaceuticals or chemotherapeutic agents were also assayed, using technetium or
Paclitaxel respectively. In conclusion, the technique allowed the production of biocompatible
and bioactive protein nanoparticles suitable for the administration of radiopharmaceuticals
and chemotherapeutic agents.

Abstract:
Crosslinked enzyme aggregates comprise more stable and highly concentrated enzymatic preparationsof current biotechnological and biomedical relevance. This work reports the development of crosslinkednanosized papain aggregates using electron beam irradiation as an alternative route for controlledenzyme crosslinking. The nanoparticles were synthesized in phosphate buffer using various ethanol con-centrations and electron beam irradiation doses. Particle size increase was monitored using dynamiclight scattering. The crosslinking formation by means of bityrosine linkages were measured by fluores-cence spectra and the enzymatic activity was monitored using Na-Benzoyl-dl-arginine p-nitroanilidehydrochloride as a substrate. The process led to crosslinked papain nanoparticles with controlled sizesranging from 6 to 11 nm depending upon the dose and ethanol concentration. The irradiation atmosphereplayed an important role in the final bioactivity of the nanoparticles, whereas argon and nitrous oxidesaturated systems were more effective than at atmospheric conditions in terms of preserving papain enzy-matic activity. Highlighted advantages of the technique include the lack of monomers and crosslinkingagents, quick processing with reduced bioactivity changes, and the possibility to be performed inside thefinal package simultaneously with sterilization.

Abstract:
Albumin is a natural drug nanocarrier as it has equivalent diameter of about 5 nm and has natural affinity for
hydrophobic and hydrophilic drugs. They can be bound in a reversible way and the delivery controlled by
endogenous mechanism. Albumin can be crosslinked by radiation alternatively to conventional systems, in
such way that dialdehydes or toxic chemicals are avoided. On the theranostic side, radioactive ions are
commonly employed for diagnostic and therapeutic applications. As an example, radioactive gold
nanoparticles are currently employed in radiotherapy whether to increase local dose deposition in tissue
during radiotherapy or as a local emitter of gamma and beta rays. The radioactive properties of gold include:
198Au (?max=0.96 MeV; t1/2 = 2.7 days) and 199Au (?max =0.46 MeV; t1/2 = 3.14 days), making it a
strong candidate for theranostics . Conjugation of such materials with sugars, peptides, antibodies, proteins
among others is routinely used nowadays for targeting. While radioactive nanoparticles can offer a much
higher dose payload than ions for therapy and diagnostic, in addition to the the huge surface to bind targeting
species presented by the nanoparticles, functionalization with proteins may potentially increases the particle
uptake by tumors or tissues. The main purpose of this work was the development of one pot in situ synthesis
of radioactive gold 198 nanoparticle encapsulated by albumin for application in cancer Theranostics. While
crosslinked albumin may provide a nontoxic coating on AuNPs with a controllable hydrodynamic diameter,
conventional AuNP can be activated by nuclear reactor to produce 198AuNP. The gamma or beta radiation
originated from the gold nanoparticle was used to crosslink the Albumin layer. The use of a radioactive
particle able to emit radiation for crosslinking of the Albumin layer and simultaneous theranostic application
was tried for the first time. The elegant procedure and ease of produc¬tion combined with the properties of
198Au and the safety of HSA-198AuNP make this new particle an exciting advancement in cancer therapy
and diagnosis. For such purpose, radioactive tetrachloroauric acid H198AuCl4 was produced from gold foils
of high purity by neutron irradiation in IPEN research nuclear reactor. 198AuNP/BSA were synthesized by
stirring aqueous solutions of BSA and radioactive tetrachloroauric acid H198AuCl4. The reaction mixtures
were stirred continuously at 25 °C. The color of the mixture become purple-red from pale yellow within 15
minutes indicating the formation of gold nanoparticles. The reaction mixture was stirred for an additional 20
minutes. The 198AuNP/BSA formed were characterized by UV-Vis spectrophotometry, DLS and TEM
analysis. The spectra were recorded at first day of preparation and after 1 month. Radioactive gold
nanoparticle encapsulated by crosslinked Albumin was prepared in reproducible way. The gold nanoparticle
core size measured by TEM was about 20 nm and about 60 to 70 nm with the albumin layer as measured by
DLS. Bityrosine formation was measured by fluorescence and it was an evidence of intramolecular and
intermolecular crosslinking. In conclusion the technique was suitable for the in situ production of the
crosslinked albumin functionalized radioactive gold nanoparticles. Acknowledgements The authors would

Abstract:
Recent studies demonstrated the development of papain and bovine serum albumin
nanoparticles using gamma radiation (10 kGy) in presence of 20-30% (v/v) ethanol.
With the purpose of producing stable and well defined nanocarriers, this work aims to
determine the influence of different dose rates over protein nanoparticle formation. For
this purpose, papain and BSA nanoparticles were synthetized in phosphate buffer (50
mM, pH 7.2) and ethanol (20-30%, v/v) using a radiation dose of 10 kGy and dose rate
of 0.8, 2, 5 and 10 kGy.h-1. After irradiation, samples were evaluated by dynamic light
scattering, fluorescence and proteolytic activity to verify the size, secondary structure
and monitoring of the enzymatic activity, respectively. For papain nanoparticles it was
observed that the dose rate did not influence the particle size formation, however
crosslinking evidenced by bityrosine showed that samples irradiated at 0.8 and 5
kGy.h-1 presented higher bityrosine levels. On the other hand, BSA nanoparticles
presented different results if compared to papain NPs. Different dose rates caused
different and non-linear size increase for each condition, following the order: 5 > 10 >
0.8 > 2 kGy.h-1. However, in terms of crosslinking formation, a linear increase was
registered, as at 0.8 kGy.h-1 the smallest signal was achieved, whereas at 10 kGy.h-
1 the highest signal was recorded. In conclusion, BSA nanoparticles were more
sensitive to different radiation dose rates than nanopapain. Optimized results in terms
of size increase and higher bityrosine levels were observed for the samples irradiated
at 5 kGy.h-1, in which nanoparticle formation will occur faster if compared to the
synthesis carried out under distinct conditions. As final applications of the system
concert their use for the delivery of chemo or radiotherapeutics, the loading of
paclitaxel, a well-known chemotherapeutic agent, and radiolabeling with tecntetium-
99m, a radioisotope suitable for biomedical applications, have also been performed
with high efficiency, thus demonstrating a proof of concept of such systems.
