Abstract:
Several diseases or conditions cause dermatological disorders that hinder the process of skin repair. The search for novel technologies has inspired the combination of stem cell (SC) and light-based therapies to ameliorate skin wound repair. Herein, we systematically revised the impact of photobiomodulation therapy (PBM) combined with SCs in animal models of skin wounds and quantitatively evaluated this effect through a meta-analysis. For inclusion, SCs should be irradiated in vitro or in vivo, before or after being implanted in animals, respectively. The search resulted in nine eligible articles, which were assessed for risk of bias. For the meta-analysis, studies were included only when PBM was applied in vivo, five regarding wound closure, and three to wound strength. Overall, a positive influence of SC + PBM on wound closure (mean difference: 9.69; 95% CI: 5.78–13.61, P < 0.00001) and strength (standardized mean difference: 1.7, 95% CI: 0.68–2.72, P = 0.001) was detected, although studies have shown moderate to high heterogeneity and a lack of information regarding some bias domains. Altogether, PBM seems to be an enabling technology able to be applied postimplantation of SCs for cutaneous regeneration. Our findings may guide future laboratory and clinical studies in hopes of offering wound care patients a better quality of life.

2016.
Abstract:

Abstract:
As células
Tipo I e colágeno tipo III, o LED de emissão vermelha apresentou maior
densidade celular.

Abstract:
λ=660nm, com 100mW entregando 3J, 4J e 20J de energia respectivamente.

Abstract:
Ionizing radiation (IR) induces DNA damage and low-level
laser therapy (LLLT) has been investigated to prevent
or repair detrimental outcomes resulting from IR exposure.
Few in vitro studies, however, explore the biological
mechanisms underlying those LLLT benefits. Thus, in this
work, fibroblasts and tumor cells are submitted to IR with
doses of 2.5 Gy and 10 Gy. After twenty-four-h, the cells
are exposed to LLLT with fluences of 30 J cm–2, 90 J cm–2,
and 150 J cm–2. Cellular viability, cell cycle phases, cell
proliferation index and senescence are evaluated on days
1 and 4 after LLLT irradiation. For fibroblasts, LLLT
promotes – in a fluence-dependent manner – increments
in cell viability and proliferation, while a reduction in the
senescence was observed. Regarding tumor cells, no influences
of LLLT on cell viability are noticed. Whereas
LLLT enhances cell populations in S and G2/M cell cycle
phases for both cellular lines, a decrease in proliferation
and increase in senescence was verified only for tumor
cells. Putting together, the results suggest that fibroblasts
and tumor cells present different responses to LLLT following
exposure to gamma-radiation, and these promising
results should stimulate further investigations.
Senescence of tumor cells and fibroblasts on the 4th day
after ionizing radiation (IR) and low-level laser therapy
(LLLT) exposures. The number of senescent cells increased
significantly for tumor cells (a) while for fibroblasts
no increment was observed (b). The blue collor indicates
senescence activity.

Abstract:

Abstract:
Candida albicans biofilm is a main cause of infections associated with medical devices such as catheters, contact lens and artificial joint prosthesis. The current treatment comprises antifungal chemotherapy thatpresents low success rates. Photodynamic inactivation (PDI) involves the combination of a photosensitiz-ing compound (PS) and light to generate oxidative stress that has demonstrated effective antimicrobialactivity against a broad-spectrum of pathogens, including C. albicans. This fungus senses glucose induc-ing an upregulation of membrane transporters that can facilitate PS uptake into the cell. The aim ofthis study was to evaluate the effects of glucose on methylene blue (MB) uptake and its influence onPDI efficiency when combined to a red LED with central wavelength at = 660 nm. C. albicans biofilmswere grown on hydrogel disks. Prior to PDI assays, MB uptake tests were performed with and withoutglucose-sensitization. In this system, the optimum PS administration was determined as 500 M of MBin contact with the biofilm during 30 min before irradiation. Irradiation was performed during 3, 6, 9, 12,15 and 18 min with irradiance of 127.3 mW/cm2. Our results showed that glucose was able to increaseMB uptake in C. albicans cells. In addition, PDI without glucose showed a higher viability reduction until6 min; after 9 min, glucose group demonstrated a significant decrease in cell viability when compared toglucose-free group. Taken together, our data suggest that glucose is capable to enhance MB uptake andmodulate photodynamic inactivation of C. albicans biofilm.

Abstract:
Over the last few years, low-level light therapy (LLLT) has shown an incredible suitability for a wide
range of applications for central nervous system (CNS) related diseases. In this therapeutic modality light
dosimetry is extremely critical so the study of light propagation through the CNS organs is of great
importance. To better understand how light intensity is delivered to the most relevant neural sites we
evaluated optical transmission through slices of rat brain point by point. We experimented red
(λ = 660 nm) and near infrared (λ = 808 nm) diode laser light analyzing the light penetration and
distribution in the whole brain. A fresh Wistar rat (Rattus novergicus) brain was cut in sagittal slices and
illuminated with a broad light beam. A high-resolution digital camera was employed to acquire data of
transmitted light. Spatial profiles of the light transmitted through the sample were obtained from the
images. Peaks and valleys in the profiles show sites where light was less or more attenuated. The peak
intensities provide information about total attenuation and the peak widths are correlated to the scattering
coefficient at that individual portion of the sample. The outcomes of this study provide remarkable
information for LLLT dose-dependent studies involving CNS and highlight the importance of LLLT
dosimetry in CNS organs for large range of applications in animal and human diseases.

Abstract:
INTRODUCTION
Photobiomodulation (PBM) has been gaining importance in
a wide range of medical fields in the past few years, particularly
in stem cell-based regenerative medicine. Improving
in vitro cell proliferation, differentiation and viability are
ways where PBM could play a pivotal role optimizing biotechnological
and bioengineering applications.
OBJECTIVES
Here we investigated whether different wavelengths (blue,
green and red) would promote distinct outcomes in human
adipose-derived stem cells (hADSCs) cultured in regular and
supplemented media for tenocyte differentiation.
MATERIALS AND METHODS
Freshly isolated hADSCs were cultured in a specific stem cell
medium (MSCGM, Lonza), DMEM or a tenogenic medium
(TEN-M: DMEM supplemented with growth factors and ascorbic
acid). Cells were irradiated every 48 h (23.28 mW/cm 2 ,
17 min 10 s delivering 24 J/cm 2 per session) using a LED irradiator
(LEDbox, BioLambda). MTT and crystal violet assays
were used to evaluate cell metabolic activity and proliferation.
DISCUSSION AND RESULTS
Red wavelength (660 nm) significantly increased metabolic
activity after five irradiations, but only for cells cultured in
TEN-M. Oppositely, blue (450 nm) and green (520 nm) light
decreased both cell proliferation and metabolic rate, with more
pronounced effects for blue light in TEN-M. Considering these
findings, we examined whether irradiating only the media would generate toxic compounds that could impair cell viability. We
therefore assessed reactive oxygen species (ROS) production
by p-nitrosodimethylaniline/histidine assay while irradiating
the three different media under the same conditions as mentioned
above. Immediately after blue and green light exposure,
an increment in ROS production was observed for DMEM and
TEN-M, that continuously increased until reaching between 4.5
and 7.1 μM one-hour after irradiation – with higher values for
TEN-M exposed to blue light.
CONCLUSION
Since no significant ROS formation was observed following
red light exposure, we concluded that medium composition
was responsible for the different effects on metabolic activity
and proliferation observed after irradiation with different
wavelengths.

Abstract:

Abstract:
Currently, antimicrobial photodynamic therapy (APDT) is limited to the local treatment of topical infections, and a platform that can deliver the photosensitizer to internal organs is highly desirable for non-local ones; SPIONs can be promising vehicles for the photosensitizer. This work reports an innovative application of methylene blue (MB)-superparamagnetic iron oxide nanoparticles (SPIONs). We report on the preparation, characterization, and application of MB-SPIONs for antimicrobial photodynamic therapy. When exposed to light, the MB photosensitizer generates reactive oxygen species (ROS), which cause irreversible damage in microbial cells. We prepare SPIONs by the co-precipitation method. We cover the nanoparticles with a double silica layer – tetraethyl orthosilicate and sodium silicate – leading to the hybrid material magnetite-silica-MB. We characterize the as-prepared SPIONs by Fourier transform infrared spectroscopy, powder X-ray diffraction, and magnetic measurements. We confirm the formation of magnetite using powder X-ray diffraction data. We use the Rietveld method to calculate the average crystallite size of magnetite as being 14 nm. Infrared spectra show characteristic bands of iron‑oxygen as well as others associated with silicate groups. At room temperature, the nanocomposites present magnetic behavior due to the magnetite core. Besides, magnetite-silica-MB can promote ROS formation. Thus, we evaluate the photodynamic activity of Fe3O4-silica-MB on Escherichia coli. Our results show the bacteria are completely eradicated following photodynamic treatment depending on the MB release time from SPIONs and energy dose. These findings encourage us to explore the use of magnetite-silica-MB to fight internal infections in preclinical assays.

Abstract:
Gold nanoshells (silica core/gold shell) have fascinating
optical extinction spectrum within visible to near infrared
range. In this work, we investigate the optical properties of
Au nanoshells and evaluate the feasibility of their use on
photodynamic therapy. Three dimensional finite element
simulation and experimental analyses were explored on
the assessment of the localized surface plasmon resonance
spectrum and spatial distribution of the electromagnetic
field enhancement near metallic nanoshells. In addition,
the interaction of Au nanoshells with methylene blue
(MB) photosensitizer was appraised, and 3.2—fold metal
enhanced single oxygen generation was observed. Lastly,
we investigate the effects of MB-mediated photoinactivation
on Staphylococcus aureus assisted by gold nanoshells.
The use of nanoshells on the photoinactivation
procedure reduced 3 the required illumination time for
total eradication of bacterial cells. Our results indicate that
Au nanoshells are promising candidates to enhance the

Abstract:
As diabetes causes millions of deaths worldwide every year, new methods for blood glucose monitoring
are in demand. Noninvasive approaches may increase patient adherence to treatment while reducing
costs, and optical coherence tomography (OCT) may be a feasible alternative to current invasive diagnostics.
This study presents two methods for blood sugar monitoring with OCT in vitro. The first, based on spatial statistics,
exploits changes in the light total attenuation coefficient caused by different concentrations of glucose in
the sample using a 930-nm commercial OCT system. The second, based on temporal analysis, calculates
differences in the decorrelation time of the speckle pattern in the OCT signal due to blood viscosity variations
with the addition of glucose with data acquired by a custom built Swept Source 1325-nm OCT system. Samples
consisted of heparinized mouse blood, phosphate buffer saline, and glucose. Additionally, further samples were
prepared by diluting mouse blood with isotonic saline solution to verify the effect of higher multiple scattering
components on the ability of the methods to differentiate glucose levels. Our results suggest a direct relationship
between glucose concentration and both decorrelation rate and attenuation coefficient, with our systems being
able to detect changes of 65 mg∕dL in glucose concentration.

Abstract:
Development of non-invasive techniques for glucose monitoring is crucial to improve glucose control and treatment adherence in patients with diabetes. Hereafter, Optical Coherence Tomography (OCT) may offer a good alternative for
portable glucometers, since it uses light to probe samples. Changes in the object of interest can alter the intensity of light
returning from the sample and, through it, one can estimate the sample's attenuation coefficient (μt) of light. In this work,
we aimed to explore the behavior of μt of mouse’s blood under increasing glucose concentrations. Different samples
were prepared in four glucose concentrations using a mixture of heparinized blood, phosphate buffer saline and glucose.
Blood glucose concentrations were measured with a blood glucometer, for reference. We have also prepared other
samples diluting the blood in isotonic saline solution to check the effect of a higher multiple-scattering component on the
ability of the technique to differentiate glucose levels based on μt. The OCT system used was a commercial Spectral
Radar OCT with 930 nm central wavelength and spectral bandwidth (FWHM) of 100 nm. The system proved to be
sensitive for all blood glucose concentrations tested, with good correlations with the obtained attenuation coefficients. A
linear tendency was observed, with an increase in attenuation with higher values of glucose. Statistical difference was
observed between all groups (p<0.001). This work opens the possibility towards a non-invasive diagnostic modality
using OCT for glycemic control, which eliminates the use of analytes and/or test strips, as in the case with commercially
available glucometers.

Abstract:
Systemic inflammation is closely related to the development of insulin resistance and type-2 diabetes, since the activation of pro-inflammatory pathways leads to inhibition of insulin signaling. Although photobiomodulation (PBM) has proven beneficial effects on the treatment of inflammatory disorders, the phototherapeutic approach to manage the chronic inflammatory component of obesity and hyperglycemia had never been explored. In this work, obese and hyperglycemic mice are treated with PBM, and their body mass, glycemia and inflammatory infiltrate of abdominal adipose tissue are evaluated. During four weeks, irradiated animals are exposed to six irradiation sessions using an 843 nm LED (5.7 J cm(-2) at 19 mW cm(-2) per session). Non-irradiated control animals display inflammatory areas almost five times greater than the treated group (p < 0.001). This result on inflammatory infiltrate may have caused impacts on the significant lower blood glucose level from irradiated animals (p = 0.04), twenty-four hours after the last irradiation session.
PBM on obese and hyperglycemic mice reduced five times the areas of inflammatory infiltrate within abdominal adipose tissue (a, b), whereas dense inflammatory regions were a common finding amidst non-irradiated animals (c). The asterisks on (c) correspond to the inflammatory infiltrate permeating adipocytes.
