Aloe vera and copaiba oleoresin-loaded chitosan films for wound dressings: microbial permeation, cytotoxicity, and in vivo proof of concept. International Journal of Pharmaceutics, v. 634, p. 1-12, 2023. DOI: 10.1016/j.ijpharm.2023.122648

Abstract: Chitosan films are commonly used for wound dressing, provided that this polymer has healing, mucoadhesiveness and antimicrobial properties. These properties can be further reinforced by the combination of chitosan with polysaccharides and glycoproteins present in aloe vera, together with copaiba oleoresin’s pharmacological activity attributed to sesquiterpenes. In this work, we developed chitosan films containing either aloe vera, copaiba oil or both, by casting technique, and evaluated their microbial permeation, antimicrobial activity, cytotoxicity, and in vivo healing potential in female adult rats. None of the developed chitosan films promoted microbial permeation, while the cytotoxicity in Balb/c 3 T3 clone A31 cell line revealed no toxicity of films produced with 2 % of chitosan and up to 1 % of aloe vera and copaiba oleoresin. Films obtained with either 0.5 % chitosan or 0.5 % copaiba oleoresin induced cell proliferation which anticipate their potential for closure of wound and for the healing process. The in vivo results confirmed that tested films (0.5 % copaiba-loaded chitosan film and 0.5 % aloe vera-loaded chitosan film) were superior to a commercial dressing film. For all tested groups, a fully formed epithelium was seen, while neoformation of vessels seemed to be greater in formulations-treated groups than those treated with the control. Our work confirms the added value of combining chitosan with aloe vera and copaiba oil in the healing process of wounds.

Antimicrobial photodynamic therapy can be an effective adjuvant for surgical wound healing in cattle. Photodiagnosis and Photodynamic Therapy, v. 33, p. 1-4, 2021. DOI: 10.1016/j.pdpdt.2020.102168

Abstract: Background Rumenostomy is a useful procedure commonly performed in cattle for medical treatment of domestic ruminants with forestomach diseases. Methylene blue (MB)-mediated antimicrobial photodynamic therapy (APDT) has been broadly investigated to treat infected wounds. Aim The aim of the study was to evaluate the effectiveness of MB-mediated APDT (MB-APDT) combined with chlorhexidine and zinc oxide ointment on wound healing process after rumenostomy. Methods Fourteen Nelore bulls were subjected to rumenostomy procedure. Animals were randomly divided into MB-APDT (MB associated with a red diode laser performed immediately after surgery and repeated on days 3, 5, 7 and 10) and control groups. Daily care included topical cleaning with chlorhexidine 2% followed by topical zinc oxide ointment. Animals were followed-up until the 28th day. Results Wounds presented a better post-surgical profile in MB-APDT group when it was compared with the control group. In MB-APDT group, it was also possible to observe less pain on palpation of wounds borders, less edema and inflammatory exudate. Additionally, animals from MB-APDT group were faster discharged from the cattle care facility. Conclusion Our results support the use of MB-APDT for the post-surgical management of rumenostomy. This pilot study ratifies the use of APDT in cattle and also suggests that it could be performed for other surgical procedures as a complementary approach or an alternative for topical administration of antibiotics.

Effect of ozone therapy on wound healing in the buccal mucosa of rats. Archives of Oral Biology, v. 119, p. 1-7, 2020. DOI: 10.1016/j.archoralbio.2020.104889

Abstract: Objective: The objective of this study was to evaluate the effects of ozone therapy on wound healing formed experimentally in the oral cavity of rats. Design: Two surgical wounds were generated on the cheeks of 24 Wistar rats, bilaterally. Half of the animals were submitted to ozone therapy on both wounds (experimental group) and the other half received no treatment (control group). In the experimental group, wounds were exposed to ozone gas 1, 2 or 3 (60 μg/mL) times. Evaluation of wound healing of the buccal mucosa was followed for 1, 3 and 7 days. The distribution of neutrophils, fibroplasia and angiogenesis were analyzed. Samples were classified in a healing numerical scale according to the inflammatory intensity. Data were submitted to Mann-Whitney and Kruskal-Wallis tests (α = 0.05). Results: On day 1, wounds were similar in both groups, lesions were open and bloody with slightly minor bleeding in the ozone therapy group. On day 3, the group with ozone therapy was almost all refurbished and with higher angiogenesis, while the control group still had more bloody points and lower blood vessels. On day 7, both wounds were remodeled, with higher fibroplasia in the group that received ozone therapy. Conclusion: It can be concluded that ozone therapy was effective in improving angiogenesis and fibroblasts count in the buccal mucosa of rats.

Microstructure and Vickers hardness of mechanically alloyed and spark plasma sintered Ti-2Zn-22Si-11B and Ti-6Zn-22Si-11B alloys. Journal of Alloys and Compounds, v. 794, p. 615-624, 2019. DOI: 10.1016/j.jallcom.2019.04.172

Abstract: This work reports on effect of zinc and milling time on microstructure and Vickers hardness of mechanically alloyed and spark plasma sintered (66,7-x)Ti-xZn-22,2Si-11,1B alloys (at.-%, with x ¼ 2 and 6 at.-%). Ball milling at different times (20, 60, 180 and 300 min) was performed under argon atmosphere in a planetary ball mill from these elemental powder mixtures. Cylinder bodies with 10mm diameter were consolidated by spark plasma sintering (1100 C, 12 min, 20MPa) under argon atmosphere. Asmilled powders and sintered Ti-Zn-Si-B samples were evaluated by X-ray diffraction, scanning electron microscopy, energy dispersive spectrometry, laser diffraction particle size analysis, and Vickers hardness tests. It was noted a similar behavior during ball milling of Ti-Zn-Si-B powder mixtures. Only the a-Ti and Si peaks were indexed in XRD patterns of Ti-Zn-Si-B powders milled for 300min. Rietveld refining indicated that the amount of amorphous structures has increased during milling up to 300 min, which was more pronounced for the Ti-6Zn-22Si-11B powders. The particle sizes of the Ti-2Zn-22Si-11B and Ti-6Zn-22Si-11B powder mixtures increased during milling from 94 (20 min) to 156 (300min) mm and from 101 (20 min) to 136 (300 min) mm, respectively. Coherently, the average specific surface area of powder particles was continuously reduced during milling. Despite the adopted parameters, the SPS process produced dense Ti-xZn-22Si-11B (x ¼ 2 and 6 at.-%) alloys with 98% from their theoretical specific mass. Regardless of milling time, the Ti6Si2B formation was inhibited in microstructure of the spark plasma sintered Ti-6Zn-22Si-11B alloy previously milled for longer times. EDS analysis indicated that the Ti6Si2B phase dissolved close to 1.8 at.-% Zn. Beside the TiB and Ti5Si3, the Ti2Zn and Ti3Zn (not explored) phases were also found in microstructure of sintered Ti-Zn-Si-B alloys. The Ti-2Zn-22Si-11B alloy presented average Vickers hardness values higher than 1050 HV whereas the Ti-6Zn-22Si-11B alloy varied between 970 and 1036 HV, which could be associated with the Ti6Si2B formation.

Abstract: Burns are one of the leading causes of morbidity worldwide and the most costly traumatic injuries. A better understanding of the molecular mechanisms in wound healing is required to accelerate tissue recovery and reduce the health economic impact. However, the standard techniques used to evaluate the biological events associated to wound repair are laborious, time-consuming, and/or require multiple assays/staining. Therefore, this study aims to evaluate the feasibility of Fourier transform infrared (FT-IR) spectroscopy to monitor the progress and healing status of burn wounds. Burn injuries were induced on Wistar rats by water vapor exposure and biopsied for further histopathological and spectroscopic evaluation at four time-points (3, 7, 14, and 21 days). Spectral data were preprocessed and compared by principal component analysis. Pairwise comparison of post-burn groups to each other revealed that metabolic activity induced by thermal injury decreases as the healing progresses. Higher amounts of carbohydrates, proteins, lipids, and nucleic acids were evidenced on days 3 and 7 compared to healthy skin and reduced amounts of these molecular structural units on days 14 and 21 postburn. FT-IR spectroscopy was used to determine the healing status of a wound based on the biochemical information retained by spectral signatures in each phase of healing. Our findings demonstrate that FT-IR spectroscopy can monitor the biological events triggered by burn trauma as well as to detect the wound status including full recovery based on the spectral changes associated to the biochemical events in each phase.

Physical, chemical, and biological characterization of biodegradable chitosan dressing for biomedical applications: could sodium bicarbonate act as a crosslinking agent?. Materials Chemistry and Physics, v. 301, p. 1-15, 2023. DOI: 10.1016/j.matchemphys.2023.127636

Abstract: Chitosan, a biomaterial with properties that allows the elaboration of biocompatible and biodegradable systems, was employed to prepare dressings - 2% (w/v) in acetic acid - followed by freezing and lyophilization. Some samples were kept chemically unchanged and used as control, while others were cross-linked with Epichlorohydrin 0.01 mol L− 1 for 24 h being washed, frozen and lyophilized. The neutralization procedure was performed with sodium bicarbonate, which ended up leading to a crosslinking by ionic interactions in the polymer network, and characteristics as promising as those of the chemically cross-linked with Epichlorohydrin. SEM showed that the crosslinking leads to a controlled formation of the polymeric network, conferring suitable steam permeation capacity 107 g mm.day− 1 .m− 2 .kPa− 1 , simulated fluid permeation between 2000 and 2500 g m− 2 .day-1 and porosity with interconnection which allows fluid absorption capacity above 900%. The crosslinking processes favored a better handling mechanical resistance confirmed by biodegradation assays. The dressing blocked micro-organisms permeation, forming an efficient barrier against contamination, is safe and biocompatible, allowing satisfactory cell adhesion and proliferation. These results showed that both dressings present statistically and satisfactory equivalent characteristics to be used as a functional bio dressing suitable for the treatment of different pathologies.

DOI: 10.1021/acsabm.8b00369

Abstract: The field of wound healing has seen an increase in research activity in wound care and hydrogel-based dressings have been targeted as a solution for these applications. Hydrogels with silver nanoparticles can present many advantages for this field. However, if the aggregation and sterilization of this product have not been carefully considered, the effectiveness or use could be limited. Therefore, in the current study, a hydrogel-based wound dressing membrane was developed using polyvinylpyrrolidone (PVP), polyethylene glycol (PEG), agar, and carboxymethyl cellulose (CMC). Silver ions (Ag+) were dispersed in the polymer matrix and its reduction with formation of a hydrogel and silver nanoparticles was performed using 60Co gamma irradiation to enhance the dressings antimicrobial properties. The resulting hydrogel presented a high degree of swelling and a good size control of silver nanoparticles. The incorporation of AgNPs was confirmed via Raman spectroscopy and the samples presented no signs of toxicity in vitro as assessed using an elution assay with neutral red uptake as the cytotoxic end point. Membranes were tested in vivo using a full thickness defeat model in rabbits. Postmortem histopathological analysis indicated that the use of the hydrogel membranes that incorporated AgNPs had a stimulatory action on wound healing as evidenced by a high intensity of fibroblasts and neovascularization in the tissue, which promoted a faster healing process when compared to the untreated wounds. We demonstrate the possibility of producing a hydrogel with good size control of AgNPs, which can also be directly sterilized within the formation of this material via gamma irradiation. Furthermore, the mechanism of hydrogel healing, in vivo, with silver nanoparticles was found to have a direct correlation of silver nanoparticles with in vitro cell results.
