Abstract:
Stingrays skin secretions are largely studied due to the human envenoming medical relevance of the sting
puncture that evolves to inflammatory events, including necrosis. Such toxic effects can be correlated to the
biochemical composition of the sting mucus, according to the literature. Fish skin plays important biological
roles, such as the control of the osmotic pressure gradient, protection against mechanical forces and microorganism
infections. The mucus, on the other hand, is a rich and complex fluid, acting on swimming, nutrition
and the innate immune system. The elasmobranch's epidermis is a tissue composed mainly by mucus secretory
cells, and marine stingrays have already been described to present secretory glands spread throughout the body.
Little is known about the biochemical composition of the stingray mucus, but recent studies have corroborated
the importance of mucus in the envenomation process. Aiming to assess the mucus composition, a new noninvasive
mucus collection method was developed that focused on peptides and proteins, and biological assays
were performed to analyze the toxic and immune activities of the Hypanus americanus mucus. Pathophysiological
characterization showed the presence of peptidases on the mucus, as well as the induction of edema and leukocyte
recruitment in mice. The fractionated mucus improved phagocytosis on macrophages and showed antimicrobial
activity against T. rubrumç. neoformans and C. albicans in vitro. The proteomic analyses showed the
presence of immune-related proteins like actin, histones, hemoglobin, and ribosomal proteins. This protein
pattern is similar to those reported for other fish mucus and stingray venoms. This is the first report depicting the
Hypanus stingray mucus composition, highlighting its biochemical composition and importance for the stingray
immune system and the possible role on the envenomation process.

Abstract:
Galectin-3 (Gal-3) controls intercellular and cell-extracellular matrix interactions during immunological
responses. In chronic inflammation, Gal-3 is associated with fibrotic events, regulates B cell
differentiation and delays lupus progression. Gal-3 deficient mice (Lgals3−/−) have intense germinal
center formation and atypical plasma cell generation correlated to high levels IgG, IgE, and IgA. Here,
we used pristane (2,6,10,14-tetramethylpentadecane) to induce lupus-like syndrome in Lgals3−/− and
Lgals3+/+ BALB/c mice. Mesentery and peritoneal cells were monitored because promptly react to
pristane injected in the peritoneal cavity. For the first time, mesenteric tissues have been associated to
the pathogenesis of experimental lupus-like syndrome. In Lgals3+/+ pristane-induced mice, mesentery
was hallmarked by intense fibrogranulomatous reaction restricted to submesothelial regions and
organized niches containing macrophages and B lymphocytes and plasma cells. In contrast, Lgals3−/−
pristane-treated mice had diffuse mesenteric fibrosis affecting submesothelium and peripheral tissues,
atypical M1/M2 macrophage polarization and significant DLL1+ cells expansion, suggesting possible
involvement of Notch/Delta pathways in the disease. Early inflammatory reaction to pristane was
characterized by significant disturbances on monocyte recruitment, macrophage differentiation and
dendritic cell (DC) responses in the peritoneal cavity of pristane-induced Lgals3−/− mice. A correlative
analysis showed that mesenteric damages in the absence of Gal-3 were directly associated with severe
portal inflammation and hepatitis. In conclusion, it has suggested that Gal-3 orchestrates histological
organization in the mesentery and prevents lupoid hepatitis in experimental lupus-like syndrome by
controlling macrophage polarization, Notch signaling pathways and DC differentiation in mesenteric
structures.

Abstract:
Gamma radiation induces protein changes that enhance immunogenicity for venoms, used in antivenin production.
Coccidian parasites exposed to gamma radiation elicit immune response with protection in mice and
man, but without studies on the effect of gamma radiation in soluble acellular extracts or isolated proteins.
Toxoplasmosis is a highly prevalent coccidian disease with only one vaccine for veterinary use but with remaining
tissue cysts. Total parasite extracts or recombinant proteins used as immunogen induce usually low
protection. Here, we study gamma radiation effect on T. gondii extracts proteins (STAG) and its induced immunity
in experimental mice models. By SDS-PAGE, protein degradation is seen at high radiation doses, but at
ideal dose (1500 Gy), there are preservation of the antigenicity and immunogenicity, detected by specific antibody
recognition or production after mice immunization. Immunization with STAG irradiated at 1500 Gy induced
significant protection in mice immunized and challenged with distinct T. gondii strains. In their blood,
higher levels of specific CD19+, CD3+CD4+ and CD3+CD8+ activated cells were found when compared to mice
immunized with STAG. Irradiated T. gondii tachyzoites extracts induce immune response and protection in mice
in addition, could be a feasible alternative for Toxoplasma vaccine.

Abstract:
This study aimed to report the effects of different doses of ionizing radiation on inflammatory and repair stage of human skin graft adherence in Nude mice wounds. Animals were divided into transplanted with irradiated human skin grafts (IHSG) at 25 and 50 kGy (IHSG 25 kGy; IHSG 50 kGy) and non-IHSG and euthanized on the 3rd, 7th and 21st days after the surgery, by gross and microscopic changes, immunostaining for human type I collagen (Col I) and mouse Col I and Col III and inflammatory cells. We found an effectiveness of human split-thickness graft adherence in mice transplanted with IHSG 25 kGy, as well decrease in dermo-epidermal necrosis and neutrophils, lower loss of skin thickness, epithelization and neo-vascularization. Day 21 post-transplantation with IHSG 25 kGy was observed a well-preserved human skin in the border of the graft, a prominent granulation tissue in an organization by proliferated fibroblasts, Col III deposition and increased B-cells and macrophages. A complete adherence of human skin graft occurred with IHSG 25 kGy. We suggest that the ionizing radiation at 25 kGy mediates inflammation and the repair stage of human skin graft adherence in murine model, thus emerging as a potential tool in healing cutaneous wounds.

Abstract:
Background: Antimicrobial peptides are present in animals, plants and microorganisms and play a fundamental
role in the innate immune response. Gomesin is a cationic antimicrobial peptide purified from haemocytes of the
spider Acanthoscurria gomesiana. It has a broad-spectrum of activity against bacteria, fungi, protozoa and tumour
cells. Candida albicans is a commensal yeast that is part of the human microbiota. However, in
immunocompromised patients, this fungus may cause skin, mucosal or systemic infections. The typical treatment
for this mycosis comprises three major categories of antifungal drugs: polyenes, azoles and echinocandins;
however cases of resistance to these drugs are frequently reported. With the emergence of microorganisms that
are resistant to conventional antibiotics, the development of alternative treatments for candidiasis is important. In
this study, we evaluate the efficacy of gomesin treatment on disseminated and vaginal candidiasis as well as its
toxicity and biodistribution.
Results: Treatment with gomesin effectively reduced Candida albicans in the kidneys, spleen, liver and vagina of
infected mice. The biodistribution of gomesin labelled with technetium-99 m showed that the peptide is captured
in the kidneys, spleen and liver. Enhanced production of TNF-a, IFN-g and IL-6 was detected in infected mice
treated with gomesin, suggesting an immunomodulatory activity. Moreover, immunosuppressed and C. albicansinfected
mice showed an increase in survival after treatment with gomesin and fluconazole. Systemic
administration of gomesin was also not toxic to the mice
Conclusions: Gomesin proved to be effective against experimental Candida albicans infection. It can be used as an
alternative therapy for candidiasis, either alone or in combination with fluconazole. Gomesin’s mechanism is not
fully understood, but we hypothesise that the peptide acts through the permeabilisation of the yeast membrane
leading to death and/or releasing the yeast antigens that trigger the host immune response against infection.
Therefore, data presented in this study reinforces the potential of gomesin as a therapeutic antifungal agent in
both humans and animals.

Para participar do RI-IPEN é necessário que pelo menos um dos autores tenha vínculo acadêmico ou funcional com o Instituto. Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.
A elaboração do projeto do RI do IPEN foi iniciado em novembro de 2013, colocado em operação interna em julho de 2014 e disponibilizado na Internet em junho de 2015. Utiliza o software livre Dspace, desenvolvido pelo Massachusetts Institute of Technology (MIT). Para descrição dos metadados adota o padrão Dublin Core. É compatível com o Protocolo de Arquivos Abertos (OAI) permitindo interoperabilidade com repositórios de âmbito nacional e internacional.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.
Operando, inicialmente como uma base de dados referencial o Repositório foi disponibilizado na atual plataforma, em junho de 2015. No Repositório está disponível o acesso ao conteúdo digital de artigos de periódicos, eventos, nacionais e internacionais, livros, capítulos, dissertações, teses e relatórios técnicos.

Para participar do RI-IPEN é necessário que pelo menos um dos autores tenha vínculo acadêmico ou funcional com o Instituto. Nesta primeira etapa de funcionamento do RI, a coleta das publicações é realizada periodicamente pela equipe da Biblioteca do IPEN, extraindo os dados das bases internacionais tais como a Web of Science, Scopus, INIS, SciElo além de verificar o Currículo Lattes. O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.
