Descriptive epidemiological and pathological study of canine renal cell carcinoma.

Abstract:
Renal cell carcinoma (RCC) accounts for 0.5–1.5% of all canine
tumor types and is very aggressive, rapidly growing, and
metastasis-generating, culminating in the death of the animal. The
management of canine RCC lacks predictive biomarkers. The aim
of this study is to conduct an epidemiological and pathological
analysis of dogs affected with RCC.A retrospective and descriptive
review of studies published between January 2001 to December
2019 was carried out using the following electronic databases: Web
of Science, PubMed, SciELO, Science Direct. Twenty-one articles
from 11 countries were included in the study, totaling 258 cases.
Histopathological analysis based on cytological criteria showed
that cromophobic and clear cell carcinoma were the most frequent
subtypes. The histological classification revealed that papillary and
tubular carcinoma accounted for 70% of the cases. Vimentin, Pax8,
CKs (CK AE1/AE3, CK CAM5.2 and CK7), COX-2, Napsin-A, CD10
and CD117 were the most commonly used diagnostic biomarkers,
corresponding to 80% of cases. Moreover, 14-3-3σ and COX-2
were also used as predictive biomarkers for canine RCC.
In conclusionVimentin, Pax8, CKs (CK AE1/AE3, CK CAM5.2
and CK7), COX-2, Napsin-A, CD10 and CD117 were the most
commonly used diagnostic biomarkers, corresponding to 80% of
cases. Moreover, 14-3-3σ and COX-2 were also used as a predictive
biomarker for canine RCC.

Abstract:
Background: Cardiovascular complications are common in chronic kidney disease (CKD) patients.
Endothelial nitric oxide synthase (eNOS) is very important for the homeostasis of the cardiovascular system,
and its gene polymorphism at position 894 (G>T) has not been investigated with QTc interval in patients on
dialysis.
Objective: This study evaluated the association of the 894G>T polymorphism with QTc prolongation and
endothelial dysfunction risk in dialysis patients.
Methods: Predialysis blood samples were collected for eNOS gene polymorphism, nitric oxide (NO),
thiobarbituric acid reactive substances (TBARS), total antioxidant, asymmetric dimethylarginine (ADMA)
and L-arginine, and 12-lead electrocardiograms were analyzed in these patients. Statistics were based on
continuous and categorical variables using Fisher’s exact or Chi-square or one-way ANOVA or Kruskal-
Wallis tests. The results were considered significant when P < 0.05.
Results: The study showed that the GG genotype was prevalent, with 54% of patients, followed by 41%
GT and 6% TT, and the genotypic distribution was not associated with QTc prolongation. Furthermore,
patients with the T allele showed increased ADMA, L-arginine and peroxidation lipid levels with reduced NO
synthesis.
Conclusion: Our study showed a lack of association between QTc interval and eNOS polymorphism; however,
it was found that patients with the T allele had a greater risk of developing endothelial dysfunction by ADMA,
which could contribute to future cardiovascular complications and worsening of CKD.

Abstract:
Acute kidney injury (AKI) is an important health
problem and can be caused by number of factors. The use of
aminoglycosides, such as gentamicin, is one of these factors.
Recently, an effort has been made to find biomarkers to guide
treatment protocols. Inductively coupled plasma optical emission
spectroscopy (ICP-OES) was used to estimate the contents
of Ca, Cu, Fe, K, Mg, Mn, Na, P, and Zn in serum and
urine of the healthy, AKI, and spontaneous recovery (SR)
groups of animals. The animal model of AKI and SR was
validated by measuring serum and urinary urea and creatinine.
The quantitative determination of the elements showed a decrease
in serum levels of Ca, and Fe in the AKI group (P<0.01
vs. healthy), with a return to normal levels in the SR group,
without a significant difference between the healthy and SR
groups. In the urine samples, there was a decrease in P and Na
levels in the AKI group (P<0.001 and P<0.01 vs. healthy), but
Ca levels were increased in this group compared with the
healthy and SR groups (P<0.01). These findings indicate that
mineral elements might be useful as biomarkers for AKI.

Despite great therapeutic advances in the last decade, metastatic RCC (mRCC) is still considered an incurable disease. In
this study, we examined the potential of essential elements as biomarkers of mRCC using an orthotropic metastatic mouse
model. Frozen lung and plasma samples from healthy and mRCC-induced mice were lyophilized, digested, and analyzed using
inductively coupled plasma mass spectrometry. In metastatic lungs, a significant increase in Ca concentration (268%) was
observed, whereas a significant decrease in Cu (23.2%), Fe (17.4%), Mn (38.8%), and Na (11.7%) was observed. The plasma
of mRCC-induced mice showed decreased concentrations of Mn (53%), Na (19.7%) and Zn (49,50%) and increased levels of
Ca (53%), Cu (39.5%), Our findings revealed marked differences in the concentrations of essential elements in the lung and
plasma of the metastatic mouse model. The circulating levels of Ca, Cu, Mn, Na, and Zn could be utilized as diagnostic and
therapeutic response biomarkers.

Abstract:
Animal models of renal disease have been used in the study of pathogenesis and therapeutic protocols. In this study, doppler ultrasound was used to evaluate dysfunction in the renal vasculature in acute kidney injury in an animal model. Eight male Wistar rats received gentamicin (80 mg/kg) for 10 days. The blood urea and creatinine levels were measured to assess renal function. All doppler ultrasound measurements were performed on both kidneys and a colour map of the renal circulation was generated. Renal function and Doppler ultrasound measurements were performed 10 days before GM treatment and on the 5th and 10th days of the assay. Gentamicin treatment led to increased serum creatinine and blood urea levels at 5 days and 10 days post initial inoculation. A significant reduction in renal artery blood flow was observed after 5 days. However, these levels remained unchanged until the 10th day, demonstrating a lack of correlation with serum creatinine and blood urea levels. Therefore, the assessment of flow blood velocity of renal arteries by doppler ultrasound is not useful for monitoring acute kidney injury in rats.

Abstract:
Prolactin (PRL) is a hormone produced by the pituitary gland with innumerable functions, such as lactation, reproduction,
osmotic and immune regulation. The present work describes the synthesis of hPRL in human embryonic
kidney (HEK293) cells, transiently transfected with the pcDNA-3.4-TOPO® vector carrying the hPRL cDNA. A concentration
of ~ 20 mg/L, including glycosylated (G-hPRL) and non-glycosylated (NG-hPRL) human prolactin, was obtained,
with ~ 19% of G-hPRL, which is higher than that observed in CHO-derived hPRL (~ 10%) and falling within the wide
range of 5–30% reported for pituitary-derived hPRL. N-Glycoprofiling analysis of G-hPRL provided: (i) identification of
each N-glycan structure and relative intensity; (ii) average N-glycan mass; (iii) molecular mass of the whole glycoprotein
and relative carbohydrate mass fraction; (iv) mass fraction of each monosaccharide. The data obtained were compared
to pituitary- and CHO-derived G-hPRL. The whole MM of HEK-derived G-hPRL, determined via MALDI–TOF-MS,
was 25,123 Da, which is 0.88% higher than pit- and 0.61% higher than CHO-derived G-hPRL. The main difference with
the latter was due to sialylation, which was ~ sevenfold lower, but slightly higher than that observed in native G-hPRL.
The “in vitro” bioactivity of HEK-G-hPRL was ~ fourfold lower than that of native G-hPRL, with which it had in common
also the number of N-glycan structures.

O RI-IPEN apresenta também um aspecto inovador no seu funcionamento. Por meio de metadados específicos ele está vinculado ao sistema de gerenciamento das atividades do Plano Diretor anual do IPEN (SIGEPI). Com o objetivo de fornecer dados numéricos para a elaboração dos indicadores da Produção Cientifica Institucional, disponibiliza uma tabela estatística registrando em tempo real a inserção de novos itens. Foi criado um metadado que contém um número único para cada integrante da comunidade científica do IPEN. Esse metadado se transformou em um filtro que ao ser acionado apresenta todos os trabalhos de um determinado autor independente das variáveis na forma de citação do seu nome.
A elaboração do projeto do RI do IPEN foi iniciado em novembro de 2013, colocado em operação interna em julho de 2014 e disponibilizado na Internet em junho de 2015. Utiliza o software livre Dspace, desenvolvido pelo Massachusetts Institute of Technology (MIT). Para descrição dos metadados adota o padrão Dublin Core. É compatível com o Protocolo de Arquivos Abertos (OAI) permitindo interoperabilidade com repositórios de âmbito nacional e internacional.

O Repositório Digital do IPEN é um equipamento institucional de acesso aberto, criado com o objetivo de reunir, preservar, disponibilizar e conferir maior visibilidade à Produção Científica publicada pelo Instituto, desde sua criação em 1956.
Operando, inicialmente como uma base de dados referencial o Repositório foi disponibilizado na atual plataforma, em junho de 2015. No Repositório está disponível o acesso ao conteúdo digital de artigos de periódicos, eventos, nacionais e internacionais, livros, capítulos, dissertações, teses e relatórios técnicos.
